A novel fusion transcription factor drives high cellulase and xylanase production on glucose in Trichoderma reesei

Summary: In order to reduce the high cost of cellulase production in lignocellulose biorefining, strategies to enhance enzyme productivity from economic and manipulatable carbon sources need to be developed. This study designed an artificial transcription factor XT by fusing the DNA binding domain of Xyr1 to the transactivation domain of Tmac1. The XT recombinant strain (OEXT) greatly improved cellulase production on repressing glucose compared with the control strain QM9414, with significant increases in pNPCase and xylanase activity.

Summary: The research discovered that a single-point mutation in the main transactivator Xyr1 of Trichoderma reesei significantly impacted xylanase expression, related to a decrease in DNA-binding affinity. The mutated Xyr1 no longer exhibited a structural change response to specific carbohydrates, suggesting the presence of a nuclear receptor-like domain that mediates carbohydrate signals and modulates its transactivating activity.

Summary: The study cloned, expressed and purified Cip1 from T. reesei, and investigated its effects on enzymatic hydrolysis of different pretreated lignocellulose. It showed that Cip1 could significantly promote the enzymatic hydrolysis, especially on substrates with high lignin content. Additionally, Cip1 could disrupt the crystal structure of cellulose and reduce non-productive adsorption of cellulase on lignin.
