Visible-Light-Mediated Modification and Manipulation of Biomacromolecules

Vivian M. Lechner et al.

Summary: Chemically modified biomacromolecules play a crucial role in chemical biology. Visible-light-induced photochemical approaches have emerged as an effective method to modify biomacromolecules, offering mild reaction conditions and precise control. These methods can be used to modify native functional groups as well as non-native, bioorthogonal functional groups, allowing manipulation of biomacromolecule activity.

Hydrogel and Effects of Crosslinking Agent on Cellulose-Based Hydrogels: A Review

Halimatuddahliana Nasution et al.

Summary: Hydrogels are polymer materials that can swell but are insoluble in water. The three-dimensional structure of hydrogels affects their properties. Hydrogels synthesized with natural polymers have better biocompatibility, while those with synthetic polymers have adjustable mechanical properties.

Protection of human γD-crystallin protein from ultraviolet C-induced aggregation by ortho-vanillin

Shu-Shun Hsueh et al.

Summary: This study investigated the effect of ortho-vanillin on ultraviolet-C-induced aggregation of human gamma D-crystallin, demonstrating its dose-dependent suppression of protein aggregation. Structural changes in gamma D-crystallin induced by the interaction with ortho-vanillin were revealed, potentially contributing to the development of therapeutics for cataracts.

Enzyme-photo-coupled catalytic systems

Shaohua Zhang et al.

Summary: Efficient chemical transformation is essential for modern society's sustainable development. Enzyme-photo-coupled catalytic systems (EPCS) show promise in solar-driven 'molecular editing', but the design of efficient and stable EPCS is challenged by the involvement of multiple components and catalytic processes. Identifying connectivity and compatibility between biocatalytic and photocatalytic ensembles as crucial factors governing EPCS performance, solutions for undesirable connectivity and low compatibility are discussed to propose designing principles for EPCS and provide a future perspective.

Engineering an acetyllysine reader with a photocrosslinking amino acid for interactome profiling

Anirban Roy et al.

Summary: The site-specific installation of light-activable crosslinker unnatural amino acids is a powerful approach to trap transient protein-protein interactions both in vitro and in vivo. In this study, the engineering of a bromodomain to introduce 4-benzoyl-L-phenylalanine using amber suppressor mutagenesis without compromising its ability to recognize acetylated histone proteins demonstrates high crosslinking efficiency towards interacting partners and suitability for profiling the transient bromodomain interactome.
