3′aQTL-atlas: an atlas of 3′UTR alternative polyadenylation quantitative trait loci across human normal tissues

Ya Cui et al.

Summary: Genome-wide association studies have identified numerous non-coding SNPs associated with human traits and diseases, with functional interpretation remaining a challenge. The concept of 3'UTR alternative polyadenylation (APA) quantitative trait loci (3'aQTLs) has been established to interpret a portion of GWAS SNPs. The 3'aQTL-atlas provides a comprehensive database of nearly 1.49 million SNPs associated with APA across 49 human tissues, aiming to explain genetic basis of APA and APA-linked susceptibility genes in human traits and diseases.

Identification of novel risk loci with shared effects on alcoholism, heroin, and methamphetamine dependence

Yan Sun et al.

Summary: This study identified and replicated three novel genetic loci associated with the common risk of heroin, methamphetamine addiction, and alcoholism. These loci have functional effects on gene expression and brain structure, and may provide insights into the root cause of substance dependence, aiding in the development of prevention strategies for addiction disorders.

Comprehensive Integrative Analyses Identify TIGD5 rs75547282 as a Risk Variant for Autism Spectrum Disorder

Xinyan Xie et al.

Summary: The study utilized a convergent functional genomics approach to identify three candidate causal genes of ASD, with the TIGD5 gene showing association with ASD risk through a specific SNP (rs75547282). Genotyping experiments revealed an increased risk of ASD associated with TIGD5 rs75547282 under the dominant model, but with limited statistical power (5.2%).

Cigarette smoking-associated isoform switching and 3′ UTR lengthening via alternative polyadenylation

Zhonghui Xu et al.

Summary: Cigarette smoking induces significant transcriptomic and systemic inflammatory responses, affecting gene expression, alternative isoform regulation, and exon usage. Lengthening of 3' UTRs associated with smoking suggests potential biomarkers and therapeutic targets for smoking-related diseases, emphasizing the importance of further research in this area.

An atlas of alternative polyadenylation quantitative trait loci contributing to complex trait and disease heritability

Lei Li et al.

Summary: The study presents a multi-tissue atlas of alternative polyadenylation (APA) quantitative trait loci (3 ' aQTLs) identifying approximately 0.4 million common genetic variants associated with the APA of target genes. About 16% of trait-associated variants colocalize with 3 ' aQTLs, indicating their substantial contribution to the molecular mechanisms behind human complex traits and diseases.

Innate immunity receptors in depression and suicide: upregulated NOD-like receptors containing pyrin (NLRPs) and hyperactive inflammasomes in the postmortem brains of people who were depressed and died by suicide

Ghanshyam N. Pandey et al.

Summary: Abnormalities in inflammation, especially in innate immunity receptors such as NLRPs and their inflammasomes, were observed in the brains of people who were depressed and died by suicide. The upregulation of NLRP1, NLRP3, NLRP6, caspase3, and ASC in this population suggests a potential role of innate immunity receptors in the pathophysiology of depression and suicide.
