Functional connectivity changes in insomnia disorder: A systematic review

Elisabetta Fasiello et al.

Summary: Insomnia is the most common sleep disorder, with unclear pathogenetic mechanisms, but studies on functional connectivity suggest disrupted brain organization may underlie symptoms. More research with shared and rigorous designs is needed to solidify understanding of insomnia pathophysiology.

Spindle-related brain activation in patients with insomnia disorder: An EEG-fMRI study

Yan Shao et al.

Summary: Sleep spindles have been related to sleep protection, depression, and anxiety. This study aimed to investigate the connection between spindle-related brain activations and sleep quality, depression, and anxiety symptoms in insomnia disorder (ID) patients. The findings suggest that decreased brain activations in the cingulate gyrus may impact subjective sleep quality and emotional regulation in individuals with ID.

Summary: Using functional magnetic resonance imaging (fMRI), researchers found that imagining fatigue-related multisensory sensations activates brain regions involved in first-person perspective, multisensory integration, and mental imagery networks.

Fatigue, Depression, and Anxiety Among Ambulating Multiple Sclerosis Patients

Safanah AlSaeed et al.

Summary: This study aims to assess the prevalence of depression, anxiety, and fatigue among pwMS who have no physical disability in Saudi Arabia and demonstrate any correlation between these factors and physical activity as well as the QoL. The results revealed that depression, anxiety, and fatigue tend to be correlated and clustered together among pwMS in our cohort.

Sleep discrepancy is associated with alterations in the salience network in patients with insomnia disorder: An EEG-fMRI study

Yuezhen Li et al.

Summary: This study found that there are sleep discrepancies in the connectivity of the anterior insula (aINS) during wake and non-rapid eye movement (NREM) sleep in patients with insomnia disorder. The connectivity of a neural pathway involving the aINS, putamen, and thalamus may underlie sleep perception.

BRAINMECHANISMS OF INSOMNIA: NEW PERSPECTIVES ON CAUSES AND CONSEQUENCES

Eus J. W. Van Someren

Summary: Insomnia is the second most common mental disorder, yet progress in understanding its underlying neurobiological mechanisms has been limited. This review explores the definition, prevalence, and susceptibility factors of insomnia, including genetic variants, early life stress, major life events, and brain structure and function. The proposed model suggests that in individuals vulnerable to developing insomnia, the locus coeruleus may be more sensitive to salience network input, leading to a spiral of hyperarousal and impaired sleep.

Altered thalamic connectivity in insomnia disorder during wakefulness and sleep

Guangyuan Zou et al.

Summary: This study investigated brain functional networks of patients with insomnia disorder and healthy controls, revealing decreased thalamic connectivity in patients with insomnia disorder compared to controls, particularly with regions like the left amygdala and parahippocampal gyrus. Specific brain region connectivity was found to be significantly correlated with sleep misperception index, shedding new light on the neuropathophysiology of insomnia disorder.

Testing the contiguity of the sleep and fatigue relationship: a daily diary study

Andrea L. Harris et al.

Summary: The study explored the relationship between sleep and fatigue, finding that self-reported nightly sleep quality significantly predicted subsequent daily fatigue ratings, while total sleep time was a significant predictor of fatigue within participants. Additionally, unhelpful sleep beliefs and rumination about fatigue mediated the relationship between insomnia and fatigue reporting.

EEG spectral analysis in insomnia disorder: A systematic review and meta-analysis

Wenrui Zhao et al.

Summary: Research shows that patients with insomnia disorder exhibit increased beta band power during wakefulness and rapid eye movement (REM) sleep, as well as decreased delta power and increased theta, alpha, and sigma power during non-rapid eye movement (NREM) sleep. The study also suggests that relative power is a more sensitive biomarker during NREM sleep compared to wakefulness and REM sleep.

Thalamocortical functional connectivity in patients with insomnia using resting-state fMRI

Nambeom Kim et al.

Summary: The study found differences in resting-state functional connectivity between patients with insomnia disorder and healthy controls, with patients showing increased hyperactivity in brain areas related to sensory functions. Some of the strengthened functional connections were negatively correlated with sleep efficiency.
