Micro-flow imaging multi-instrument evaluation for sub-visible particle detection

Summary: Sub-visible particles (SVPs) in pharmaceutical products are critical quality attributes and should be monitored during development. Micro-Flow Imaging (MFI) is a flow imaging method that overcomes the shortcomings of light obscuration (LO) in detecting smaller translucent SVPs. In this study, seven MFI instruments were evaluated for counting and size precision and accuracy, and the results were compared to alternative methods, such as LO and Backgrounded Membrane Imaging (BMI). The precision and accuracy of particle count and size were found to depend on the particle source and material properties, and limitations were observed in the detection limits claimed by the manufacturer. Additionally, proteinaceous particles showed significant differences in particle counts, while particle diameters were consistent across sizes.

Afraid of the wall of death? Considerations on monoclonal antibody characteristics that trigger aggregation during peristaltic pumping

Summary: Protein aggregation is a major concern in the manufacturing of biopharmaceuticals. It is triggered by protein adsorption to tubing surfaces and subsequent film rupture during peristaltic pumping for filtration, transfer, or filling. Our study found that the affinity of proteins to the surface, hydrophobicity, and conformational stability play important roles in aggregation propensity.

Random Heteropolymer Excipients Improve the Colloidal Stability of a Monoclonal Antibody for Subcutaneous Administration

Rapid Insights into the Stabilization of Interferon Alpha by Two Surfactants Revealed by STD-NMR Spectroscopy

Summary: This study demonstrates the application of saturation transfer difference (STD) nuclear magnetic resonance (NMR) spectroscopy in detecting protein-surfactant interactions. It was found that polysorbate 20 binds with interferon alpha, with stronger binding at pH closer to the protein's iso-electric point. In contrast, no interactions were detected between poloxamer 407 and interferon alpha. This research highlights the potential of STD-NMR for quickly assessing surfactant-protein interactions and aiding in surfactant selection for protein formulation.

Proteins on the Rack: Mechanistic Studies on Protein Particle Formation During Peristaltic Pumping

Summary: The need for filtering intravenous infusions has long been recognized, even though scientific evidence on the indications for in-line filters is limited. Recent papers and clinical studies have highlighted the importance of defining the type of filtration, its potential benefits, and its proper indications. The WoCoVA Foundation initiated a consensus project on intravenous filtration, aiming to raise global awareness on the risks of intravenous access and patient safety. A panel of experts was assembled to summarize current knowledge on filtration and guide future research in the field.

Catching Speedy Gonzales: Driving forces for Protein Film Formation on Silicone Rubber Tubing During Pumping

Summary: Interfacial adsorption and particle formation during the processing of biopharmaceutics were studied in this research. The driving forces and rate of adsorption of a monoclonal antibody to the silicone rubber surface, as well as the influence of formulation parameters on particle concentration and size distribution, were investigated. It was found that formulation parameters, pH, and ionic strength, had an effect on the total amount of adsorbed protein and the formation of particles larger than 25 mm.
