Formation of a Stable Co-Amorphous System for a Brick Dust Molecule by Utilizing Sodium Taurocholate with High Glass Transition Temperature

Summary: This study aimed to formulate a stable co-amorphous system for brick dust molecules by utilizing sodium taurocholate (NaTC) with a higher glass transition temperature (Tg). Ball milling formed a co-amorphous system for AntiY(5)R and NaTC (AntiY(5)R-NaTC) at various molar ratios. The results showed that AntiY(5)R-NaTC had an 8.5-fold increase in solubility compared to its crystalline form, and the co-amorphous system remained amorphous for 1 month at 25°C and 40°C.

Insight into the mechanism behind oral bioavailability-enhancement by nanosuspensions through combined dissolution/permeation studies

Summary: As numerous new drug candidates are poorly water soluble, enabling formulations are needed to increase their bioavailability for oral administration. This study aimed to gain insight into nanoparticle characteristics and performance using an in vitro combined dissolution/permeation setup. The results showed that nanosuspensions exhibited significantly increased dissolution rates and permeation rates compared to the raw APIs.

Poly (amino acid)s as new co-formers in amorphous solid dispersion

Summary: The drug-amino acid co-amorphous systems and amorphous solid dispersions (ASDs) are promising methods for poorly water-soluble drugs. Poly (amino acid)-based ASDs with high drug-loading and physical stability were prepared by ball milling poly-L-lysine and polyglutamic acid with mebendazole, tadalafil, and valsartan. The dissolution rate and physical stability of the drugs were improved by the drug-poly (amino acid) ASDs. Salt formation between the drug and poly (amino acid) is not necessary for the formation of promising ASDs.

Summary: The majority of new drugs have poor water solubility, which requires the development of enabling formulations for sufficient bioavailability upon oral administration. This study compared the biopredictive abilities of two in vitro tools for predicting intestinal supersaturation and systemic absorption of drug formulations. The PermeaLoop™ apparatus showed better predictive ability compared to the mu FLUX™ setup. The study confirms the usefulness of these tools for formulation ranking of weakly basic compounds and supersaturating formulations.

Sole-amorphous-sugar-based solid dispersion of curcumin and the influence of formulation composition and heat treatment on the dissolution of curcumin
