3D printed, personalized sustained release cortisol for patients with adrenal insufficiency

S. Ayyoubi et al.

Summary: The standard of care for patients with Adrenal Insufficiency (AI) needs improvement by adopting a personalized approach. A novel 24 h sustained release 3D printed (3DP) hydrocortisone formulation (M3DICORT) has been developed to mimic physiological cortisol plasma concentration, with uniform drug distribution and stable dissolution profile. Impurities in the final product need to be addressed before clinical assessment.

3D printing for personalised medicines: implications for policy and practice

Klaudia Englezos et al.

Summary: The healthcare dynamic has shifted to patient-centred care, requiring more individualised therapies. However, the pharmaceutical industry has not fully adapted to this shift, preventing pharmacists from providing completely personalised medicine to patients. This article reviews the limitations of current pharmaceutical manufacturing methods, discusses the benefits of 3D printing for personalised medicine, and explores the implications for pharmacy practice and policy.

Towards point-of-care manufacturing and analysis of immediate-release 3D printed hydrocortisone tablets for the treatment of congenital adrenal hyperplasia

Tzuyi L. Yang et al.

Summary: This study developed immediate-release hydrocortisone tablets using FDM 3D printing and assessed drug contents using near-infrared spectroscopy. The results demonstrated the feasibility of this method for determining drug content. Integration of non-destructive process analytical technology can accelerate the adoption of individualised dosing in a clinical setting.

Fused deposition modeling 3D printing of solid oral dosage forms containing amorphous solid dispersions: How to elucidate drug dissolution mechanisms through surface spectral analysis techniques?

Chloe Parulski et al.

Summary: This study aims to enhance the dissolution rate of poorly soluble drugs using hot-melt extrusion coupled with fused deposition modeling. The results demonstrated that solid oral forms containing challenging drugs can be successfully prepared using hot-melt extrusion and 3D printing. The amorphous state of the drug showed improved dissolution rate, and the rate was influenced by the infill density and polymer composition of the printed forms.

Adrenal insufficiency

Eystein S. Husebye et al.

Summary: Adrenal insufficiency can be caused by various factors such as primary adrenal disorder, adrenocorticotropic hormone deficiency, or suppression of adrenocorticotropic hormone by certain medications. Clinical features include unintentional weight loss, anorexia, hypotension, fatigue, muscle and abdominal pain, and hyponatremia. Diagnosis is often delayed due to non-specific symptoms, and patient education on managing the condition is crucial to prevent adrenal crisis.

3D printed spherical mini-tablets: Geometry versus composition effects in controlling dissolution from personalised solid dosage forms

Sejad Ayyoubi et al.

Summary: Oral dosage forms are widely used but can cause adverse effects due to standardized mass produced solid dosage forms. 3D printing allows for personalized medicine, as shown in the study focusing on hypertensive patients using nifedipine. The drug dissolution is more impacted by composition rather than geometry and surface area.

Can filaments be stored as a shelf-item for on-demand manufacturing of oral 3D printed tablets? An initial stability assessment

Tochukwu C. Okwuosa et al.

Summary: 3D printing of oral solid dosage forms is a newly introduced approach for personalized dosing, with Fused Deposition Modelling (FDM) being a promising technique. The success of FDM relies heavily on the mass manufacturing of stable filaments. The study investigated the stability of various polymer filaments over 6 months and their impact on the potential of on-demand manufacturing of 3D printed tablets.

Influence of active pharmaceutical ingredient structures on Hansen solubility parameters

Eva Petrikova et al.

Summary: Preformulation is a crucial step in drug development, where the compatibility between API and excipients is determined by Hansen solubility parameters (HSPs) which can also describe the surface properties of substances. In this study, the HSPs of six selected APIs with different chemical nature were determined using inverse gas chromatography (IGC), revealing the influence of API structure on HSPs and the sensitivity of the method to different chemical substances. This provides valuable insights into API behavior during drug development.
