Swelling of Homogeneous Alginate Gels with Multi-Stimuli Sensitivity

Haniyeh Malektaj et al.

Summary: A new two-step method for preparing homogeneous alginate gels is proposed. In the first step, alginate chains are weakly bonded by Ca2+ ions in a low-pH aqueous solution. In the next step, the gel is immersed in a strong solution of CaCl2 to complete the cross-linking process. Homogeneous alginate gels retain their integrity in aqueous solutions with pH ranging from 2 to 7, ionic strength from 0 to 0.2 M, and temperatures from room temperature up to 50 degrees C, and can be used in biomedical applications.

Global trend in exosome isolation and application: an update concept in management of diseases

Mohammadhassan Omrani et al.

Summary: Extracellular vesicles (EVs) have great potential for disease diagnosis and treatment. Among EVs, exosomes are a subtype of spherical vesicles with a lipid layer, released from various cells and involved in signal transduction and homeostasis. However, existing methods for isolating and characterizing EVs and exosomes have limitations. New approaches in EV biology show promise in better understanding their biological function. This review discusses the strengths and limitations of conventional strategies and novel microfluidic methods for EV isolation, as well as commercially available exosome isolation techniques and kits and the global market demand for exosome assays.

Interplay Between Extracellular Matrix Remodeling and Angiogenesis in Tumor Ecosystem

Rohit Siddhartha et al.

Summary: Studying tumorigenesis mechanisms and neoplastic cell interactions in the tumor ecosystem is crucial for effective cancer treatment. The dynamic tumor ecosystem consists of tumor cells, extracellular matrix (ECM), secreted factors, and various cell types. ECM remodeling and release of growth factors promote endothelial cell proliferation, migration, and angiogenesis. Stromal cancer-associated fibroblasts (CAF) release angiogenic cues that interact with ECM proteins, enhancing angiogenesis and supporting tumor growth. Targeting angiogenesis and ECM components is a major strategy for anticancer treatment. Exploring these agents in a context-specific manner may overcome therapy resistance and reduce tumor burden.

Robin Augustine et al.

Summary: The lack of oxygen supply in engineered constructs poses a major challenge in tissue engineering and regenerative medicine. Providing and maintaining adequate oxygen levels in large tissue-engineered constructs as well as during transportation and storage is crucial. Recent approaches involve incorporating oxygen-generating materials such as peroxides and fluorinated substances into scaffolds. This review paper highlights the importance of oxygen-generating materials in various tissue engineering approaches and discusses challenges, solutions, and future strategies in designing clinically relevant multifunctional biomaterials.

Mechanical Properties of Alginate Hydrogels Cross-Linked with Multivalent Cations

Haniyeh Malektaj et al.

Summary: This study investigates the preparation of cross-linked alginate hydrogels using various cations and analyzes their equilibrium swelling and mechanical properties. The results show that the type of cations used in the cross-linking process affects the elastic moduli and equilibrium degree of swelling. Experimental data is fitted with a model that involves two material parameters, and numerical analysis reveals the influence of cations on these quantities. The study also finds that the relationship between the elastic modulus and equilibrium swelling differs from that predicted by conventional theory for covalently cross-linked gels.

Pro-angiogenic Potential of Mesenchymal Stromal Cells Regulated by Matrix Stiffness and Anisotropy Mimicking Right Ventricles

Michael Nguyen-Truong et al.

Summary: This study assessed the impact of right ventricle (RV) tissue mechanics on mesenchymal stromal cell (MSC) behavior. It was found that MSCs produced the highest levels of vascular endothelial growth factor (VEGF) in the failing, isotropic group, which was greater than the normal, isotropic group (p < 0.05). The presence of anisotropy led to trends of increased VEGF production in normal groups, but this effect was absent in failing groups.

Summary: Myocardial infarction, the leading cause of heart failure worldwide, has sparked interest in stem cell therapy for cardiac repair. While some successful results have been achieved in animal models, the overall outcome of clinical trials has been disappointing, mainly due to limited stem cell survival and retention. Combinational use of stem cells and various priming strategies may hold promise for improving regenerative cell therapy efficacy.
