Summary: Alzheimer's Disease (AD) disproportionately affects women and individuals with the APOE-epsilon 4 susceptibility gene. This study aimed to understand the influence of these two risk factors on brain atrophy in AD and healthy aging. The results showed that women experience more severe brain atrophy in the later stages of the disease, and the APOE genotype accelerates brain atrophy in AD patients. These findings have important implications for targeted interventions.

Self-reported cognitive decline among middle and older age autistic adults

Summary: Very little is known about the cognitive decline in aging autistic adults. However, this study found a high rate of cognitive decline among middle and older age autistic adults, with symptoms including declining interest in leisure activities and problems with thinking, memory, and judgment. Autistic individuals designated female at birth may be more vulnerable to cognitive decline than those designated male at birth. This study highlights the potential utility of self-report measures for early screening of cognitive decline in autistic adults as they age.

Summary: Alzheimer's disease is the most common cause of dementia worldwide, and the APOE gene is the strongest and most prevalent genetic risk factor. The ε4 allele increases AD risk while the ε2 allele is protective. ApoE protein has multiple functions, including lipid transport and promoting the formation of amyloid-beta plaques. Different apoE isoforms have different effects on Aβ-related pathways. The complexity of apoE biology presents challenges to developing effective therapeutic strategies.

Effects of age on the hippocampus and verbal memory in adults with autism spectrum disorder: Longitudinal versus cross-sectional findings

Summary: Research on aging in adults with autism spectrum disorder (ASD) is increasing, but longitudinal studies are needed. This study found that middle-aged adults with ASD have greater hippocampal volume loss and more clinically meaningful decline in short-term memory compared to neurotypical (NT) adults. In contrast, cross-sectional analyses showed that increasing age was associated with worsening short-term memory in NT adults, but not in autistic adults. Reduced integrity of connections to the hippocampus and greater challenges with long-term memory were also found in autistic adults across a broad age range. These preliminary longitudinal findings highlight the importance of studying aging in autistic adults over time.
