Mast cell tolerance in the skin microenvironment to commensal bacteria is controlled by fibroblasts

Summary: This study investigates the activation and degranulation mechanism of mast cells, as well as the interaction between mast cells and dermal fibroblasts in reducing unnecessary inflammation when exposed to beneficial commensal bacteria. The study reveals that the extracellular matrix hyaluronic acid functions as an activator for the regulatory proteins A20/TNFAIP3, which reduces the mast cells' response to commensal bacteria. The anti-inflammatory role of hyaluronic acid presents new possibilities for the treatment of inflammatory and allergic disorders.

Summary: Hematopoietic stem cells (HSCs) have the ability to self-renew and differentiate into all mature hematopoietic cell types. They are mainly found in the bone marrow and are regulated by a hierarchy of different types of cells. Mast cells, a type of innate immune cell, have a complex developmental process starting from the yolk sac and later originating from HSCs. This review article summarizes the knowledge on mast cell sources and the mechanisms involved in their development.
