Conformational analysis and interaction of the Staphylococcus aureus transmembrane peptidase AgrB with its AgrD propeptide substrate

Summary: The virulence gene expression in S. aureus is controlled by the agr quorum sensing system, and the AgrB and AgrD proteins play important roles in this process. Through homology modelling and molecular dynamics techniques, the conformations of AgrB and AgrD were characterized and their interaction mechanisms were analyzed. Experimental evidence confirmed the direct interaction between AgrB and AgrD, and the formation of AgrBD complex in vitro. These findings contribute to a better understanding of the mechanisms behind the regulation of virulence gene expression in S. aureus.

Summary: Continuous rotation of liquid bacterial culture in a random positioning machine (RPM) leads to the formation of a colloidal bacterial culture in the culture tube, altering the microenvironment of bacterial cells in simulated microgravity. This change in environment may induce changes in cell wall and surface properties, affecting growth kinetics, cell wall constitution, surface hydrophobicity, autoaggregation ability, and antibiotic susceptibility of Staphylococcus aureus NCIM 2079 strain. Noteworthy alterations were observed, especially in susceptibility to Erythromycin and Clindamycin, suggesting that microgravity may cause changes in the cellular envelope of planktonic S. aureus cultures.
