Journey to the centre of the lung. The perspective of a mineralogist on the carcinogenic effects of mineral fibres in the lungs

Gasdermin D membrane pores orchestrate IL-1α secretion from necrotic macrophages after NFS-rich silica exposure

Summary: IL-1 alpha is released by macrophages and contributes to silica-induced lung inflammation. The molecular mechanism of IL-1 alpha release from particle-exposed macrophages is unclear. This study found that only reactive silica particles with a high content of nearly free silanols could trigger IL-1 alpha release through the formation of GSDMD pores in the membrane.

Non-asbestiform elongate mineral particles and mesothelioma risk: Human and experimental evidence

The session also covered the National Mesothelioma Virtual Bank, the consensus on the potential of long, durable asbestiform EMPs to cause mesothelioma, and experimental studies on engineered nanomaterials.

Detection of fine asbestos fibers using fluorescently labeled asbestos-binding proteins in talc

Summary: In this study, we evaluated the feasibility of using fluorescence microscopy (FM) for detecting fine asbestos fibers in powdered talc. FM showed superior sensitivity in detecting nano-scale fibers and enabled the selective visualization of asbestos fibers, even when hidden beneath numerous talc particles. A semi-quantitative analysis of asbestos contamination in talc was achieved using a point counting method with FM images.

Gasdermin D mediates the maturation and release of IL-1α downstream of inflammasomes

Summary: This study demonstrates that GSDMD is essential for the rapid maturation of IL-1α by nonparticulate inflammasome activators. Ablation of GSDMD prevents the maturation of IL-1α but not IL-1β. The maturation of IL-1α induced by inflammasomes relies on extracellular calcium and calpains, which are induced in a GSDMD-dependent manner.
