Action spectra and mechanisms of (in) efficiency of bipolar electric pulses at electroporation

Summary: This study aimed to achieve the maximum cancellation of electroporation by quantifying the impact of pulse shape, duration, number, and repetition rate on a broad range of electric field strengths. The addition of an opposite polarity phase reduced cell membrane permeabilization, with the strongest cancellation achieved by adding a 50% second phase to 600-ns pulses delivered at a high repetition rate. Strobe photography revealed that the temporal summation of polarization enhanced electroporation for unipolar pulses, while no summation was observed for bipolar pulses.

Next generation CANCAN focusing for remote stimulation by nanosecond electric pulses

Summary: By canceling the low efficiency of bipolar nanosecond electric pulses, we introduced a new remote focusing method that effectively weakens the electric field with distance. The new method utilizes unipolar nsEP packets to produce bipolar waveforms near the electrodes, suppressing electroporation, but not at the remote target. In experiments, we observed 1.5-2 times stronger electroporation in the center of a quadrupole electrode array compared to near the electrodes, despite a 3-4-fold field attenuation. When the array was lifted above the cell monolayer, simulating a 3D treatment, the remote effect was enhanced up to 6-fold.
