Emerging Hallmarks of Metabolic Reprogramming in Prostate Cancer

Francesco Lasorsa et al.

Summary: Prostate cancer is a common malignancy in men and a leading cause of cancer death. The cancer cells have specific metabolic characteristics, including increased lipogenesis, cholesterogenesis, and reliance on glutamine. The tumor microenvironment also plays a role in cancer progression through interactions between cancer and stromal cells. The use of multi-omic approaches can provide a comprehensive understanding of the metabolic changes in prostate cancer.

The cross-talk between Abl2 tyrosine kinase and TGFβ1 signalling modulates the invasion of clear cell Renal Cell Carcinoma cells

Sofia De Marco et al.

Summary: In this study, it was found that TGF-beta 1/Smad signaling is more activated in high-grade ccRCC and inversely correlated with Abl2 protein expression. TGF-beta 1 treatment increased the degradation of Abl2 protein and promoted tumor invasion and metastasis markers in ccRCC cells.

Radiogenomics in Renal Cancer Management-Current Evidence and Future Prospects

Matteo Ferro et al.

Summary: Renal cancer management poses challenges throughout the entire process from diagnosis to treatment and follow-up. The differentiation between benign and malignant tissues in cases of small renal masses and cystic lesions can be problematic, even with imaging or renal biopsy. Recent advances in artificial intelligence, imaging techniques, and genomics provide potential for helping clinicians with risk stratification, treatment selection, follow-up strategies, and prognosis. However, further prospective studies with larger patient cohorts are needed to validate previous results and implement these techniques into clinical practice.

MUC1 Expression Affects the Immunoflogosis in Renal Cell Carcinoma Microenvironment through Complement System Activation and Immune Infiltrate Modulation

Giuseppe Lucarelli et al.

Summary: MUC1 is overexpressed in clear cell renal cell carcinoma (ccRCC) and is associated with aggressive cancer phenotype and chemoresistance. This study found that MUC1 can regulate the immunoflammatory response in the tumor microenvironment by activating the classical pathway of the complement system and is associated with the expression of pentraxin-3 (PTX3). The results suggest that MUC1 expression can modulate the immune infiltration in the ccRCC microenvironment and promote the formation of an immune-silent microenvironment.

Cellular and Molecular Players in the Tumor Microenvironment of Renal Cell Carcinoma

Francesco Lasorsa et al.

Summary: Globally, clear-cell renal cell carcinoma (ccRCC) is the most prevalent type of kidney cancer. Surgery is key in treatment, but a significant number of patients have metastatic ccRCC or experience recurrence. Molecular-target-based agents like TKIs and ICIs are recommended for advanced cancer treatment. The tumor microenvironment (TME) plays an essential role in cancer development and can be targeted for therapy. Understanding immune cell interactions with cancer and other TME elements is crucial for enhancing immunotherapies and reducing resistance.

Summary: The American Cancer Society predicts that there will be 1,958,310 new cancer cases and 609,820 cancer deaths in the United States in 2023. While incidence trends for most cancers are favorable, prostate cancer saw a significant increase of 3% annually from 2014 to 2019. However, the overall cancer death rate continues to decline due to advances in treatment, with a 33% reduction since 1991.

Understanding and Overcoming Immunosuppression Shaped by Cancer Stem Cells

Linheng Li et al.

Summary: The use of immunotherapy has revolutionized cancer treatment for certain types of cancers, but its broad utility is limited due to ineffectiveness in many cancer types. This is partly due to the phenotypic plasticity established by cancer stem cells (CSC) and their associated microenvironments, which allows for immune evasion. This article discusses the current understanding of CSC-mediated immune evasion and proposes ways to overcome the immune privilege of CSCs and the immune-suppressive microenvironment.

Novel Insights into Autophagy and Prostate Cancer: A Comprehensive Review

Davide Loizzo et al.

Summary: Autophagy plays a pivotal role in the regulation of apoptosis and disease progression in prostate cancer. However, depending on the cellular context, it can have either detrimental or protective effects. Current research focuses on modulating autophagy to improve therapeutic response and chemotherapy resistance in prostate cancer, and identifying the genetic signature of autophagy as a potential means to assess aggressiveness.

Cell plasticity, senescence, and quiescence in cancer stem cells: Biological and therapeutic implications

Ritama Paul et al.

Summary: Cancer stem cells (CSCs) play a crucial role in cancer progression, metastasis, relapse, and therapy resistance. They maintain their self-renewal capability and resist the tumor microenvironment and treatments through mechanisms such as cellular plasticity, senescence, and quiescence. Understanding these mechanisms is important for future therapeutic implications.

Pharmacological disruption of the MTDH-SND1 complex enhances tumor antigen presentation and synergizes with anti-PD-1 therapy in metastatic breast cancer

Minhong Shen et al.

Summary: By targeting the MTDH-SND1 interaction through genetic and pharmacological approaches, it has been found that the complex plays a crucial role in suppressing antitumor T cell responses in breast cancer, ultimately enhancing immune surveillance and sensitivity to anti-programmed cell death protein 1 therapy. This suggests that combination therapy could be a viable approach to increase immune-checkpoint blockade therapy responses in metastatic breast cancer, providing a potential new strategy for treatment.
