Suppression of fatty acid oxidation supports pancreatic cancer growth and survival under hypoxic conditions through autophagy induction

Byungjoo Kim et al.

Summary: Hypoxia induces autophagy in PDAC cells, potentially through the suppression of fatty acid oxidation (FAO), which is crucial for their growth and survival. Reduction in cellular acetyl-CoA levels caused by FAO inhibition leads to a decrease in LC3 acetylation and subsequent autophagosome formation. Targeting FAO may provide a promising therapeutic approach for PDAC treatment by reducing hypoxia-induced autophagy and increasing cell death.

KEGG for taxonomy-based analysis of pathways and genomes

Minoru Kanehisa et al.

Summary: KEGG is a manually curated database resource that integrates various biological objects classified into systems, genomic, chemical, and health information. Through the KEGG identifier and URL links, users can retrieve and browse information for different biological objects, including pathway maps, hierarchy, and genomes. KEGG has also improved KO assignment annotation and included more eukaryotic genomes for better representation of organisms in the taxonomic tree. The taxonomy mapping enables analysis of the conservation of functional and physical links among genes in different organism groups.

Biomarkers for pancreatic cancer based on tissue and serum metabolomics analysis in a multicenter study

Rui Zhao et al.

Summary: This study identified metabolic features of pancreatic ductal adenocarcinoma (PDAC) and discovered potential early detection biomarkers for PDAC through tissue and serum metabolomics analysis. The identified biomarker panel showed promising performance in distinguishing PDAC from benign pancreatic neoplasms and healthy controls, especially when combined with carbohydrate antigen 19-9 (CA19-9).

Cancer-associated fibroblasts in pancreatic ductal adenocarcinoma

Tianyi Zhang et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is a deadly cancer with a high deposition of extracellular matrix (ECM) and poor prognosis. ECM proteins derived from tumor cells reduce the effectiveness of conventional cancer treatment and contribute to tumor progression and metastasis. Cancer-associated fibroblasts (CAFs) in the ECM are promising targets for novel anti-tumor interventions.

Therapeutic Potential of Adipose Stem Cell-Derived Conditioned Medium on Scar Contraction Model

Yukiko Imai et al.

Summary: By analyzing gene expression and histological morphology, we found that the patient-derived keloid model reflects scar contraction. Additionally, conditioned media derived from adipose stem cells (ASC-CM) were found to suppress scar contracture by inhibiting inflammation-related gene expression in the early phase and promoting extracellular matrix-related gene expression in the later phase.

Cancer-Associated Fibroblasts in Pancreatic Ductal Adenocarcinoma: An Update on Heterogeneity and Therapeutic Targeting

Utpreksha Vaish et al.

Summary: PDAC is a major cause of cancer-related mortality in the western world, with CAFs playing a key role in the tumor microenvironment and being potential targets for therapy. However, there is significant heterogeneity in CAFs within PDAC, highlighting the importance of understanding and delineating this heterogeneity before targeting them for treatment.

Adipose-derived mesenchymal stem cells differentiate into heterogeneous cancer-associated fibroblasts in a stroma-rich xenograft model

Yoshihiro Miyazaki et al.

Summary: Cancer-associated fibroblasts (CAFs) play important roles in tumor progression and drug resistance in pancreatic ductal adenocarcinoma (PDAC), but existing mouse models have limitations in reproducing the characteristics of clinical CAFs. Researchers have developed a new human cell-derived stroma-rich CDX model, which successfully recapitulates the clinical features of pancreatic cancer by co-transplanting human adipose-derived mesenchymal stem cells and a PDAC cell line into mice.

Role of glutamine and its metabolite ammonia in crosstalk of cancer-associated fibroblasts and cancer cells

Xiao Li et al.

Summary: CAF, the most abundant cells in the tumor microenvironment, plays a crucial role in cancer initiation, progression, metastasis, and metabolism. GLN, as a nitrogen reservoir for cancer cells and ammonia as a byproduct of its metabolism, have potential therapeutic implications in the crosstalk between CAFs and cancer cells.

Metabolic reprogramming of cancer-associated fibroblasts and its effect on cancer cell reprogramming

Zhenzhen Li et al.

Summary: Cancer cells adapt their metabolism to proliferate and survive in harsh environments, with a close relationship between tumor microenvironment and cancer-associated fibroblasts (CAFs) playing key roles in tumor growth and metastasis. CAFs act as major regulators in shaping tumor metabolism, especially through dysregulation of metabolic pathways, influencing cancer cell behavior and response to therapy. The interaction and crosstalk between cancer cells and CAFs contribute to metabolic reprogramming that impacts cancer cell growth and progression.

Metabolites and the tumour microenvironment: from cellular mechanisms to systemic metabolism

Ilaria Elia et al.

Summary: The article provides a detailed examination of the complexity of cellular metabolism within the tumor microenvironment, along with potential therapeutic intervention opportunities. Cancer cell metabolism and behavior are regulated by multiple factors, and the metabolic environment within the TME is influenced by various tiers. Understanding the interactions of these tiers reveals unique therapeutic potential for modulating the metabolic profile and function of all cells in the TME.
