Despite the availability of software and databases, the identification of compounds is still challenging. Isotope labeling has been proven as a useful tool for small molecule identification, and in this study, we introduce a novel approach using biocatalytic insertion of oxygen-18 isotope to discover and annotate previously unknown metabolites. This method, combined with high-resolution mass spectrometry and advanced data processing techniques, enhances confidence in interpreting metabolism data.

16O/18O-exchange internal standard preparation enhancing reliability of bio-sample natural bioactive compounds absolute quantitation

This study optimized a 16O/18O exchange reaction for preparing stable and affordable 18O-labeled standards. The use of 18O-labeled internal standards improved the accuracy and precision of NBCs quantitation in pharmacokinetics analysis, providing a reliable strategy for pharmaceutical research with NBCs.

Untargeted Lipidomics after D2O Administration Reveals the Turnover Rate of Individual Lipids in Various Organs of Living Organisms

Summary: The administration of low doses of D2O has been used for decades to investigate metabolic pathways and measure compound turnover rate. Previously, the uptake of deuterium in lipids was mainly studied using GC-MS instruments, but recent untargeted LC-MS/MS lipidomics approaches have been employed. In this study, the deuterium uptake in over 500 lipids was investigated in 13 organs and body fluids of mice after 4 days of 100% D2O administration. The highest deuteration was observed in the liver, plasma, and lung, while lower levels were found in the brain and heart. The incorporation of deuterium in palmitic and stearic fragments in lipids was demonstrated using MS/MS, but not in the corresponding free forms. The results were analyzed based on lipid metabolic pathways.

Characterization of free oligosaccharides from garden cress seed aqueous exudate using PGC LC-MS/MS and NMR spectroscopy

Summary: The presence of free oligosaccharides, mainly hexose containing oligosaccharides, such as raffinose, stachyose, and verbascose, was found in cress seed aqueous exudate. In addition, minor fractions of planteose, isomeric tri- and tetrasaccharides were also observed. Structural confirmation of the abundant tri- and tetrasaccharide was obtained through NMR analysis. Therefore, the presence of oligosaccharides in cress seeds enhances its bio-functionalities.

Amine additives for improved in-ESI H/D exchange

This study investigated the use of amines as modifiers to improve the extent of exchange. The extent of exchange was found to be in the order of primary amines > secondary amines > tertiary amines. These findings have implications for improving H/D exchange applications.
