Summary: In higher eukaryotes, the regulation of developmental gene expression is determined by enhancers, which can be located far away from the promoters they regulate. The architecture of chromosomes and the mechanisms of enhancer-promoter interaction play a crucial role in the development of organisms. Mammals and Drosophila share key architectural proteins and similar mechanisms in chromosome organization. This review discusses the current progress in understanding the formation and regulation of long-range interactions between enhancers and promoters in Drosophila.

Topological screen identifies hundreds of Cp190-and CTCF-dependent Drosophila chromatin insulator elements

Summary: Drosophila insulators are DNA elements that regulate gene expression by limiting chromatin contacts. By mapping chromatin contacts in Drosophila cells without key insulator proteins CTCF and Cp190, we discovered hundreds of insulator elements. Our findings suggest that Drosophila insulators play a minor role in overall genome folding but have a local impact on chromatin contacts. Our insulator catalog is an important resource for studying genome folding mechanisms.

The Mediator complex regulates enhancer-promoter interactions

They found that depletion of Mediator led to a reduction in enhancer-promoter interaction frequencies and gene expression, as well as an increase in interactions between CTCF-binding sites. These changes in chromatin architecture were associated with a redistribution of the Cohesin complex on chromatin and a decreased Cohesin occupancy at enhancers. Overall, this study provides insights into the molecular mechanisms of communication between enhancers and promoters.

A small molecule BCL6 inhibitor as chemosensitizers in acute myeloid leukemia

Summary: The spatial organization of chromosomes plays a crucial role in regulating various intranuclear processes, including gene expression in higher eukaryotes. Drosophila has been used as a model to study gene expression regulation, leading to the discovery of many transcription factors that are homologous to those involved in gene expression regulation in mammals. The current understanding suggests that the architecture of mammalian chromosomes is primarily determined by a cohesin complex, which forms chromatin loops on anchors created by CTCF, a DNA-binding architectural protein. However, the role of the cohesin complex in chromosome architecture in Drosophila is not well understood, and CTCF is just one of many architectural proteins in Drosophila that have the potential to organize specific long-range interactions between regulatory elements in the genome. This review compares the mechanisms responsible for long-range interactions and chromosome architecture between mammals and Drosophila.
