Summary: This study reveals that OPN3 is an important photoreceptor in human epidermal keratinocytes, and it is critical for the formation of supranuclear cap mediated by UVA. OPN3 mediates the formation of supranuclear cap through the calcium-dependent G protein-coupled receptor signaling pathway, which ultimately upregulates the expression of Dync1i1 and DCTN1 in human epidermal keratinocytes by activating calcium/CaMKII, CREB, and Akt signal transduction. These findings greatly expand our understanding of the phototransduction mechanisms involved in physiological function in skin keratinocytes.

Reconstructed human pigmented skin/epidermis models achieve epidermal pigmentation through melanocore transfer

Summary: The skin acts as a protective barrier against environmental insults and prevents DNA damage through the production and dispersion of melanin in the epidermal layer. Melanin transfer involves complex interactions between melanocytes, keratinocytes, and fibroblasts. By studying a newly developed human pigmented skin model, we provide evidence that melanin is transferred through coupled exocytosis/phagocytosis, similar to previous observations in human skin biopsies. These findings are valuable for understanding skin pigmentation mechanisms and disorders, and reduce the need for animal models and human skin biopsies.

Despite having a lower incidence, melanoma is the most aggressive form of skin cancer and responsible for the majority of related deaths. While it provides protection against UV radiation, it also leads to DNA damage and promotes tumor growth. Melanin secretion regulates the invasiveness of melanoma cells and affects the tumor microenvironment and immune responses.

Three-dimensional structure analysis of melanocytes and keratinocytes in senile lentigo

Summary: Senile lentigo, also known as age spots, are hyperpigmented macules of the skin caused by long-term exposure to UV radiation. The accumulation of melanosomes produced by melanocytes in neighboring keratinocytes is thought to be responsible. Different microscopic modalities were used to reconstruct the epidermal microstructure, revealing the complex interactions between melanocytes and keratinocytes in senile lentigo. The localization of melanosomes in dendritic processes surrounding recipient keratinocytes contributes to efficient melanosome transfer in senile lentigo.

Marliolide Derivative Induces Melanosome Degradation via Nrf2/p62-Mediated Autophagy

Summary: This study demonstrated that the marliolide derivative DMF02 can induce melanosome degradation through autophagy pathway by activating Nrf2, leading to reduced melanin pigmentation. Additionally, DMF02 was effective in reducing UVB-induced hyperpigmentation and regulating melanosome degradation via suppression of p62. These findings suggest that Nrf2 activators could be promising therapeutic agents for reducing hyperpigmentation.
