Single-cell transcriptomics reveal extracellular vesicles secretion with a cardiomyocyte proteostasis signature during pathological remodeling

Eric Schoger et al.

Summary: In a study on the hypertrophy of cardiomyocytes, it was found that aberrant Wnt activation leads to a proteostasis signature in extracellular vesicles and transcriptomic profiling, indicating early patho-physiological states. Aberrant Wnt activation has been observed in failing cardiomyocytes. Further analysis shows enrichment of extracellular vesicles related processes in hearts with Wnt activation, and the identification of differentially enriched proteins including proteasome constituents, chaperones, and cardiac proteins. These findings suggest that the secretion of extracellular vesicles with a proteostasis signature plays a role in the early adaptation of cardiomyocytes and may have diagnostic and prognostic implications.

Cardioprotective efficacy of limb remote ischaemic preconditioning in rats: discrepancy between a meta-analysis and a three-centre in vivo study

Nabil Sayour et al.

Summary: In this study, RIPC was found to have no cardioprotective effect in rats, based on randomized and blinded in vivo studies in three study centres using different RIPC protocols. However, a systematic review and meta-analysis of in vivo rat models showed that RIPC could reduce infarct size by an average of 21.28%. These results highlight the need for more well-designed and well-reported studies to confirm these findings and develop clinically translatable cardioprotective interventions.

Global burden of heart failure: a comprehensive and updated review of epidemiology

Gianluigi Savarese et al.

Summary: Heart Failure (HF) is a multi-faceted and life-threatening syndrome with significant morbidity and mortality worldwide, affecting over 64 million people. While the incidence of HF has stabilized in industrialized countries, the prevalence is increasing due to population ageing and improved treatment. Developing countries lack substantial data on HF epidemiology, which shows different features compared to the Western world. This review provides updated estimates on global burden of HF including prevalence, incidence, outcomes, and costs.

PACAP-38 and PAC1 Receptor Alterations in Plasma and Cardiac Tissue Samples of Heart Failure Patients

Dora Szabo et al.

Summary: This study aimed to investigate the presence of PACAP-38 in heart failure (HF) patients and its correlation with HF predictors. The results showed that plasma PACAP-38 immunoreactivity was higher in acute HF patients, while lower in chronic HF patients. In addition, a positive correlation between plasma PACAP-38 and NT-proBNP levels was observed in acute HF, but a negative correlation in chronic HF. The levels of cytokines IL-1 beta, IL-2, IL-4 were lower in chronic HF, and IL-10 was higher in acute HF. PACAP-38 levels in myocardial tissues were lower in all end-stage HF patients. The study suggests that PACAP-38 may be a potential prognostic biomarker in HF.

Rosiglitazone Does Not Show Major Hidden Cardiotoxicity in Models of Ischemia/Reperfusion but Abolishes Ischemic Preconditioning-Induced Antiarrhythmic Effects in Rats In Vivo

Bennet Y. Weber et al.

Summary: The chronic administration of rosiglitazone does not have significant hidden cardiotoxic effects in myocardial ischemia/reperfusion injury models. However, it may inhibit the antiarrhythmic effects of ischemic preconditioning, which requires further exploration.

Single-nucleus profiling of human dilated and hypertrophic cardiomyopathy

Mark Chaffin et al.

Summary: The study identified extensive molecular alterations in failing hearts through single-nucleus RNA sequencing, including changes in transcriptional profiles and the presence of a unique population of activated fibroblasts that are almost absent from non-failing samples. The findings provide new insights into the transcriptional diversity of the human heart in health and disease, as well as potential therapeutic targets and biomarkers for heart failure.

Montelukast Use Decreases Cardiovascular Events in Asthmatics

Malvina Hoxha et al.

Summary: The observational retrospective study suggests that the use of montelukast may be protective against incident cardiac ischemic events in older asthmatic patients. Analyses indicate that exposure to montelukast remains a significant protective factor for incident ischemic events, leading to a considerable risk reduction. The event-free Kaplan-Meier survival curves confirm the lower cardiovascular event incidence in patients exposed to montelukast.
