Outer Membrane Porins Contribute to Antimicrobial Resistance in Gram-Negative Bacteria

Gang Zhou et al.

Summary: This review examines the properties and interactions of five common outer membrane proteins (Omps), including OmpA, OmpC, OmpF, OmpW, and OmpX, in relation to porin-mediated permeability in gram-negative bacteria. It also highlights the discovered regulatory characteristics and identified molecular mechanisms in antibiotic penetration through Omps. Uncovering the functional properties of Omps will pave the way for investigating effective agents or approaches that use Omps as targets to eliminate resistant gram-negative bacteria.

Salmonella-Based Biorodenticides: Past Applications and Current Contradictions

Anton E. Shikov et al.

Summary: The idea of using pathogens, such as Salmonella, to control pests has been around since the late 19th century. Salmonella strains have been used as rodenticides due to their host restriction, but were banned in many countries in the second half of the 20th century due to their potential pathogenicity for animals and humans. Recent research has revealed the complexity of the molecular mechanisms affecting the host specificity and restriction of Salmonella.

Expression of membrane beta-barrel protein in E. coli at low temperatures: Structure of Yersinia pseudotuberculosis OmpF porin inclusion bodies

Tamara F. Solov'eva et al.

Summary: The study investigates the low growth temperature effect on the structure of inclusion bodies (IBs) produced by the recombinant OmpF porin of Yersinia pseudotuberculosis. The results show that introducing a N-terminal hexahistidine tag into the porin molecule increases protein biosynthesis at low cultivation temperatures. The properties and structural organization of IBs, as well as the solubilization of the porin from IBs, were studied using various techniques. Amyloid-like structures were also found in the studied IBs.

RopB protein of Rhizobium leguminosarum bv. viciae adopts amyloid state during symbiotic interactions with pea (Pisum sativum L.)

Anastasiia O. Kosolapova et al.

Summary: In this study, we found that Rhizobium bacteroids in pea nodules produce extracellular RopB amyloids, and pea nodules also contain aggregates of pea vicilin amyloid protein. By conducting in vitro experiments, we demonstrated that vicilin amyloid protein can cross-seed RopB fibrillogenesis.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.

β-Barrels and Amyloids: Structural Transitions, Biological Functions, and Pathogenesis

Anna Sulatskaya et al.

Summary: Both amyloids and beta-barrel proteins have beta-sheet-rich structures, with the latter being able to form functional amyloids in vivo. These beta-barrel amyloid proteins can interact with each other and form toxic oligomers, potentially contributing to the development of amyloidoses. Rapidly growing discoveries suggest that the number and diversity of functions of amyloid-forming beta-barrel proteins are significantly greater than currently understood.

The food additive fast green FCF inhibits α-synuclein aggregation, disassembles mature fibrils and protects against amyloid-induced neurotoxicity

Fenghua Wang et al.

Summary: The food additive fast green FCF was found to inhibit alpha-synuclein fibrillogenesis and reduce cytotoxicity by disrupting the pentamer structure, indicating its potential as a drug candidate against amyloid fibril aggregation in Parkinson's disease. The interaction mechanism between fast green FCF and alpha-synuclein was elucidated through molecular dynamics simulations, revealing the disruption of pentamer structure and reduction of beta-sheet content. Electrostatic interactions, hydrogen bonds, and pi-pi interactions were identified as key factors in the binding of fast green FCF to alpha-synuclein.

Modulation of amyloid fibrillation of bovine β-lactoglobulin by selective methionine oxidation

Sanhita Maity et al.

Summary: Oxidation-modified proteins deposition during normal aging and oxidative stress are associated with systemic amyloidoses. Met oxidation plays a crucial role in protein structure and aggregation, particularly in whey protein, affecting their properties in dairy industry. Studies indicate that oxidized beta-lg shows enhanced structural flexibility and aggregation propensity, with possible cytotoxic effects. A molecular docking study predicts selective oxidation of methionine residues at specific positions and suggests a new route for oxidative stress proteins in neurological disorders.
