Selectively Fluorinated PAMAM-Arginine Conjugates as Gene Delivery Vectors

Summary: In this study, we successfully functionalized PAMAM G2 and PAMAM G4 with fluorinated arginine-based building blocks, resulting in a novel PAMAM-based gene delivery vector. The conjugates showed improved binding capacity to plasmid DNA, reduced cytotoxicity, and enhanced gene transfection efficiency compared to undecorated PAMAM dendrimers. These findings are of great importance for gene delivery research and also have the potential for future application in F-19 magnetic resonance imaging.

The Synthesis and Transfection Activity of Disulfide Polycationic Amphiphiles

Summary: Novel polycationic amphiphiles with a disulfide group were synthesized. Cationic liposomes formed by these compounds and a helper lipid 1,2-dioleoyl-sn-glycero-3-phosphatidylethanolamine showed low cytotoxicity and efficient delivery of a fluorescently labeled oligodeoxyribonucleotide in HEK293 and HeLa cells. The efficacy of plasmid DNA delivery varied depending on the cell line and the structure of the amphiphiles, with liposomes based on tetracationic amphiphiles being the most effective transfectants. These liposomes can be useful for in vitro transfection of eukaryotic cells and further in vivo biological studies.
