Photobiology of lipofuscin granules in the retinal pigment epithelium cells of the eye: norm, pathology, age

Summary: Lipofuscin granules (LGs) accumulate in the retinal pigment epithelium (RPE) cells and can cause pathology and accelerate aging. The review examines the composition, spectral properties, and photoactivity of LGs. Studies show that the fluorescent characteristics of LGs change in pathology and after exposure to ionizing radiation. Improved methods of early diagnosis of degenerative diseases can be developed by analyzing the fluorescence spectra and kinetics of oxidized products of LG fluorophores.

These molecules, known as bisretinoids, are generated as a byproduct of vitamin A aldehyde activity and can cause reactive oxygen species, leading to photo-oxidation and photolysis. Bisretinoids accumulate in RPE cells with age and are implicated in retinal disease.
