Miswired Proprioception in Amyotrophic Lateral Sclerosis in Relation to Pain Sensation (and in Delayed Onset Muscle Soreness)-Is Piezo2 Channelopathy a Principal Transcription Activator in Proprioceptive Terminals Besides Being the Potential Primary Damage?

Balazs Sonkodi

Summary: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by progressive loss of motoneurons. The cause of ALS is largely unknown, but it is believed that both genetic mutations and aging contribute to the development of the disease. A recent theory suggests that irreversible degeneration of proprioceptive terminals leads to progressive impairment of the proprioceptive circuitry, resulting in motoneuron loss and eventually death. Additionally, Piezo2 channelopathy may play a significant role in the pathogenesis of ALS.

Mechanoreceptor Piezo1 Is Downregulated in Multiple Sclerosis Brain and Is Involved in the Maturation and Migration of Oligodendrocytes in vitro

Maria Velasco-Estevez et al.

Summary: In this study, the expression and role of the mechanoreceptor Piezo1 in oligodendrocytes and oligodendrocyte progenitor cells in the human brain were investigated. It was found that Piezo1 inhibition increased the proliferation and migration of MO3.13 oligodendrocytes, while activation of Piezo1 had opposite effects. Moreover, lower expression of Piezo1 was observed in the white matter of multiple sclerosis brains compared to healthy controls. This suggests that Piezo1-induced signaling may play an important role in the pathophysiology of multiple sclerosis.

Osteoporosis in Light of a New Mechanism Theory of Delayed Onset Muscle Soreness and Non-Contact Anterior Cruciate Ligament Injury

Balazs Sonkodi et al.

Summary: Osteoporosis is a disorder with an unknown pathomechanism, which may result from chronic overloading of the Piezo2 ion channel, leading to re-injury and early aging. The study suggests that delayed muscle soreness, non-contact anterior cruciate injury, and osteoporosis could share a common mechanism involving the Piezo2 channelopathy, but with different outcomes due to environmental factors and genetic predisposition.

Psoriasis, Is It a Microdamage of Our Sixth Sense? A Neurocentric View

Balazs Sonkodi

Summary: Psoriasis is a complex systemic disease with unclear pathomechanism. This article suggests that the microinjury of somatosensory terminals, specifically the Piezo2 channelopathy, may play a crucial role in the development of psoriasis. The primary injury occurs in the Merkel cell-neurite complex, leading to impaired glutamate release and dysregulated keratinocyte proliferation and differentiation. Additionally, the leakage of mtHsp70 from damaged mitochondria through somatosensory terminals is proposed to initiate autoimmune and autoinflammatory processes in psoriasis.

Should We Void Lactate in the Pathophysiology of Delayed Onset Muscle Soreness? Not So Fast! Let's See a Neurocentric View!

Balazs Sonkodi

Summary: The pathophysiology of delayed onset muscle soreness is not fully understood. Lactic acid, previously believed to be responsible, is now thought to have a smaller role. However, it appears that lactate plays an important role in the initiation of muscle soreness within the intrafusal space and can contribute to secondary damage in the extrafusal space by potentiating the role of bradykinin.

CD3+/CD56+ NKT-like Cells Show Imbalanced Control Immediately after Exercise in Delayed-Onset Muscle Soreness

Balazs Sonkodi et al.

Summary: The purpose of this study was to investigate the immunophenotypical characterization, with a focus on natural killer cells, of junior swimmers from the Hungarian National Swim Team before and after intense exercise. The findings showed an increase in the percentage of CD3(-)/CD56(+) natural killer cells and CD3(-)/CD56(dim+) NK cells, as well as a decrease in the percentage of CD3(+) T cells among lymphocytes after exercise. Additionally, a link between delayed-onset muscle soreness and imbalanced control of CD3(+)/CD56(+) NKT-like cells among lymphocytes was demonstrated for the first time.

Kv1.3 inhibition attenuates neuroinflammation through disruption of microglial calcium signaling

Alla F. Fomina et al.

Summary: In the past 5 years, inhibitors of the potassium channel K(V)1.3 have been shown to reduce neuroinflammation in various disease models by reducing microglia activation and suppressing pro-inflammatory cytokines and nitric oxide production. Recent studies have revealed that K(V)1.3 channels not only regulate membrane potential, but also play a crucial role in regulating calcium influx in microglia, similar to their role in T cells.
