Epigenetic Silencing of P-Element Reporter Genes Induced by Transcriptionally Active Domains of Constitutive Heterochromatin in Drosophila melanogaster

Summary: Reporter genes inserted into the Drosophila genome have been used to monitor chromatin domains and it is commonly thought that these genes are silenced when inserted into heterochromatin. However, research has found that heterochromatin is genetically and molecularly heterogeneous and contains functional genes. This study provides evidence that even when integrated within or near actively transcribed loci in the pericentric regions of chromosome 2, reporter genes are strongly silenced. Some reporter genes were found to be insensitive to a known suppressor of position effect variegation (PEV), indicating different epigenetic regulators or mechanisms control different regions of heterochromatin.

Summary: The Drosophila melanogaster beta 2-tubulin gene has been evaluated for use in biocontrol strategies for the spotted-wing Drosophila. By isolating and studying the ortholog Ds-beta 2t from the USA laboratory strain of D. suzukii, researchers have confirmed its potential for sexing and mating confirmation. Transgenic strains expressing a fluorescent protein under the control of the Ds-beta 2t promoter showed efficient sexing capabilities and comparable survival rates to wild-type males.

Summary: The spotted-wing Drosophila is originally from eastern Asia, but has now become a global threat to fruit production. Through the establishment of genetic editing techniques, Cas9-expressing strains of this species have been successfully generated and characterized. These strains show high efficiency and temperature dependence, making them useful for genetic studies in D. suzukii.

A transgenic female killing system for the genetic control of Drosophila suzukii

Summary: A new conditional female-killing (FK) strategy based on the tetracycline-off system was developed to combat the spotted wing Drosophila. Through testing different genetic constructs and transgenic strains, a construct that achieved female-specific lethality in the absence of tetracycline was identified, effectively controlling the population of female pests. Suggestions for potential improvements to the gene constructs and the use of FK strains in a sterile insect technique (SIT) program are discussed.
