Novel amino analogs of the trimethoxyphenyl ring in potent colchicine site ligands improve solubility by the masked polar group incorporation (MPGI) strategy

Myriam Gonzalez et al.

Summary: We have designed new A ring analogs with chameleonic masked polar amino groups to increase aqueous solubility and behave as non-polar through intramolecular hydrogen bonds when bound to tubulin. Some of these compounds showed improved aqueous solubility and nanomolar anti-proliferative potencies against human cancer cell lines, with the most favorable substituent being a 3-methylamino group. The antiproliferative effect is related to tubulin inhibition and cell cycle arrest in G2/M, leading to apoptosis.

Tumor Microenvironment and Glioblastoma Cell Interplay as Promoters of Therapeutic Resistance

Edoardo Agosti et al.

Summary: Despite advancements in molecular discoveries and surgical technologies, the prognosis for glioblastoma (GBM) remains poor. Tumor recurrence, particularly of GBM, could be attributed to the persistence of a subset of cancer cells with stem cell characteristics. The tumor microenvironment (TME) and its interaction with GBM stem cells have been identified as crucial factors in recurrence, making them potential targets for new therapeutic approaches.

An update on the recent advances and discovery of novel tubulin colchicine binding inhibitors

Haoxiang Weng et al.

Summary: Microtubules are a major target for inhibiting tumor cell proliferation, and microtubule-targeted agents have become increasingly effective in cancer treatment. Colchicine binding site inhibitors (CBSIs) have a simpler chemical structure than taxane and vinblastine, making them easier to modify. Furthermore, CBSIs have a strong antiproliferative effect on multidrug-resistant tumor cells and have become the mainstream research focus in the development of microtubule-targeted agents.
