A Pathogenic Role for FcγRI in the Immune Response against Chlamydial Respiratory Infection

Jiajia Zeng et al.

Summary: Fc gamma RI, an important cell surface receptor, is found to regulate host resistance against chlamydial invasion during C. muridarum respiratory infection. The absence of Fc gamma RI leads to reduced chlamydial loads and pulmonary pathology. Increased Fc gamma RI expression is observed in various immune cell populations, and Th1 cells and macrophages are identified as its target cells. The Th1 response is enhanced while M1 macrophage polarization is constrained in Fcgr1(-/-) mice, suggesting the potential role of Fc gamma RI in regulating host immunity and the inflammatory response during chlamydial infection.

IL-27 Signaling Promotes Th1 Response by Downregulating IL-10 Production in DCs during Chlamydial Respiratory Infection

Jiajia Zeng et al.

Summary: IL-27/IL-27R signaling plays a protective role in Chlamydia trachomatis infection by promoting Th1 response and suppressing neutrophilic inflammation. IL-27 expression was observed in dendritic cells (DCs), and its signaling in DCs restricted the production of IL-10, thus supporting the Th1 response against chlamydial infection.

IL-21 impairs pro-inflammatory activity of M1-like macrophages exerting anti-inflammatory effects on rheumatoid arthritis

Leilei Jian et al.

Summary: IL-21 inhibits LPS-mediated cytokine production in rheumatoid arthritis synovial fluid macrophages and impairs the pro-inflammatory activity of M1-like macrophages, exerting anti-inflammatory effects on RA. Therefore, IL-21 may not be an appropriate therapeutic target for RA.

Impact of IL-21-associated peripheral and brain crosstalk on the Alzheimer's disease neuropathology

Sudhanshu Agrawal et al.

Summary: This study reveals the role of IL-21 in Alzheimer's disease (AD) pathogenesis. IL-21 enhances peripheral and brain immune and inflammatory responses, leading to increased deposition of amyloid beta (Aβ) plaques in AD. The expression of IL-21 receptor (IL-21R) is elevated in both peripheral and brain tissues of AD patients and mice.

The Role of Macrophages in Staphylococcus aureus Infection

Grace R. Pidwill et al.

Summary: Staphylococcus aureus, a member of human commensal microflora, can cause serious diseases by circumventing the innate immune system and establishing infections. Professional phagocytes like macrophages and neutrophils play key roles in containing and resolving S. aureus infections, while the bacteria have evolved multiple strategies to survive, manipulate, and escape from macrophages. Understanding the interactions between macrophages and S. aureus may be beneficial for future therapeutic developments.

Wenhao Niu et al.

Summary: The IL-21/IL-21R interaction is involved in host defense against C. muridarum lung infection, with IL-21R(-/-) mice exhibiting a stronger CD4(+) T cell response and higher Th1/Th17 cytokine levels, leading to reduced bacterial burden and milder lung pathology. Conversely, administration of recombinant mouse IL-21 worsens chlamydial lung infection by inhibiting Th1 and Th17 responses.

Summary: The combination of MPLA and IFNγ can polarize tumor-associated macrophages to kill cancer cells, reducing primary tumor growth and metastasis. This approach also enhances chemotherapy response and suppresses metastasis, with both macrophages and T cells playing critical roles in the anti-metastatic effects. Engaging a systemic anti-tumor immune response through activation of macrophages and T cells may represent a promising strategy for cancer treatment.
