Summary: Mangosteen, also known as the queen of fruits, is a tropical fruit that has gained popularity due to its health-promoting attributes. It has been used in traditional medicine to treat various illnesses and has been shown to exhibit numerous biological and pharmacological activities, including antioxidant, anti-inflammatory, antibacterial, antifungal, antimalarial, antidiabetic, and anticancer properties. The anticancer potential of mangosteen constituents, particularly the xanthones, is currently being studied.

Novel xanthone antibacterials: Semi-synthesis, biological evaluation, and the action mechanisms

Summary: In this study, twenty-one alpha-MG derivatives were designed, synthesized, and evaluated for their antibacterial activities. The structure-activity relationships (SARs) showed that the phenolic hydroxyl groups of alpha-MG contribute to its antibacterial activity in the order of C3 > C6 > C1, with the C3 position being essential. Among the derivatives, 10a with an acetyl group at C1 exhibited higher selectivity, no hemolysis, and stronger antibacterial efficacy. Mechanistic studies revealed that 10a depolarized membrane potentials, leading to increased leakage of bacterial proteins. Our findings provide valuable insights for the development of alpha-MG-based antibacterial agents through structural modifications at C1.

Xanthones and Phenylpropanoids from the Whole Herb of Swertia pseudochinensis and Their Anti-Inflammatory Activity

Summary: An undescribed xanthone dimer, along with eleven known compounds, was isolated from the dried whole herb of Swertia pseudochinensis. Compounds 8-12 were isolated from the Swertia genus for the first time. The structure of the xanthone dimer was determined through chemical evidence and spectral analysis, and its inhibitory effects on NO production in LPS-induced RAW 264.7 cells were tested. Compound 12 showed significant inhibition, with an IC50 value of 3.05 +/- 1.10 mu M. It was hypothesized that compound 12 may bind neutrophil elastase to exert its anti-inflammatory effects through target screening and molecular docking.

Erythrocyte-derived liposomes for the treatment of inflammatory diseases

Summary: Effective and safe therapies to counteract persistent inflammation are necessary. Erythrocyte-derived liposomes (EDLs) containing omega-3 fatty acids and the anti-inflammatory drug diclofenac were developed. The EDLs were functionalised with folic acid for active targeting of inflammatory macrophages. The EDLs were able to reduce pro-inflammatory cytokine levels, comparable to dexamethasone, demonstrating their potential as bioactive carriers for inflammatory diseases.

Chiral derivatives of xanthones and benzophenones: Synthesis, enantioseparation, molecular docking, and tumor cell growth inhibition studies
