Statistical improvement of protease production from a new isolate Bacillus thuringiensis strain-MA8 and its application in the production of enzyme-modified cheese

Summary: Enzyme-modified cheese (EMC) is a concentrated cheese flavor produced enzymatically from dairy substrates. In this study, protease production conditions were optimized to increase EMC production by 7-fold. Addition of protease extracts improved the flavor and texture of EMC. Chemical analysis showed increased volatile fatty acids, water-soluble nitrogen, and acidity in EMC. The amino acid profile of EMC1 treated with protease had the highest essential amino acids content. Nutritional parameters of EMC were higher than control. Scanning Electron Microscopy revealed significant changes in EMC compared to the control.

Multimodal approaches for the improvement of the cellular folding of a recombinant iron regulatory protein in E. coli

Summary: This study investigates how the modulation of physiological stimuli and process conditions can increase the solubility and activity of recombinant proteins in E. coli cells. The results show that the presence of exogenous molecular chaperones and osmolytes, as well as the modulation of temperature and inducer concentrations, can enhance the folding of soluble aggregates and improve the functional protein molecules in the cells. The co-expression of GroEL/ES not only aids in protein folding, but also reduces cellular stress caused by aggregation-prone recombinant protein expression.

Compatibility and Washing Performance of Compound Protease Detergent

Summary: Protease is an important component of detergents, and its combination with additives can enhance the washing effect. This experiment used various methods to evaluate the whiteness and stain residues on fabric samples before and after washing. A protease detergent formula with superior decontamination and anti-deposition effects was selected. The combination of alkaline protease, keratinase, and trypsin showed cost-effective stain removal. In addition, the composite protease exhibited better overall decontamination than commercial detergents when used with suitable surfactants, anti-redeposition agents, and water-softening agents.

Regulation of heterologous subtilin production in Bacillus subtilis W168

Summary: In this study, the model organism B. subtilis W168 was successfully established as a novel platform for subtilin biosynthesis, and the underlying regulatory mechanism was comprehensively characterized. This work not only facilitates genetic (engineering) studies on subtilin, but also paves the way for its industrial production. Moreover, it sheds new light on the heterologous production of other lantibiotics.

Proteins from Thermophilic Thermus thermophilus Often Do Not Fold Correctly in a Mesophilic Expression System Such as Escherichia coli

Summary: Majority of protein structure studies use model organisms like E. coli as expression systems. However, differences in cellular environment factors, particularly temperature and chaperones, can cause improper folding of native proteins in extremophiles. This study compares the effects of using E. coli as expression system on protein structures between mesophilic bacteria and a thermophile T. thermophilus, and finds significant differences, suggesting the need for unique protein expression systems in protein folding studies of extremophiles.

Summary: In this study, a heterologous expression, enzymatic characterization, and structure homology modeling of a subtilisin-like alkaline serine protease (ASP) from Bacillus halodurans C-125 were reported. The recombinantly produced ASP demonstrated remarkable pH stability, notable thermostability, and high specific activity at extreme alkaline conditions, making it a potential novel protease candidate for various industrial sectors such as textile, detergent, leather, feed, waste, pharmaceutical, and others.
