Melatonin Can Modulate Neurodegenerative Diseases by Regulating Endoplasmic Reticulum Stress

Yeong-Min Yoo et al.

Summary: As people age, their risks of developing degenerative diseases such as cancer, diabetes, Parkinson's Disease (PD), Alzheimer's Disease (AD), rheumatoid arthritis, and osteoporosis increase. Neurodegenerative diseases are recognized as the leading cause of affliction in the elderly worldwide. Research suggests that endoplasmic reticulum (ER) stress is associated with some neurological diseases like PD and AD. Melatonin, a hormone synthesized in the pineal gland, has various biological functions including circadian rhythm control, immune enhancement, and antioxidant, anti-aging, and anti-tumor effects. This paper aims to summarize the effects of melatonin on neurons and neurodegenerative diseases related to ER stress, highlighting its role in protecting neurons and treating neurodegeneration.

Role of melatonin receptor 1B gene polymorphism and its effect on the regulation of glucose transport in gestational diabetes mellitus

Lijie Wei et al.

Summary: This study investigated the relationship between the rs10830963 variant and gestational diabetes mellitus (GDM) development, as well as the effects of the MT2 receptor on glucose uptake and transport in trophoblasts. The results showed that the genotype and allele frequencies of rs10830963 were significantly different between GDM and normal pregnant women. Additionally, melatonin could stimulate glucose uptake, GLUT4 and PPAR gamma protein expression in trophoblasts, which could be attenuated by MT2 receptor knockdown.

Agomelatine: a potential novel approach for the treatment of memory disorder in neurodegenerative disease

Qiang Su et al.

Summary: Agomelatine is a medication used to treat major depressive episodes in adults by acting as a selective agonist of melatonin receptor 1A/melatonin receptor 1B and antagonist of 5-hydroxytryptamine 2C receptors. It reconstructs sleep-wake rhythms and normalizes circadian disturbances, as well as enhances the release of dopamine and noradrenaline, reducing depression and anxiety disorder. The combination of these effects makes agomelatine unique in the treatment of depression, anxiety, and circadian rhythm disorders.

Oxidative Stress and Aging as Risk Factors for Alzheimer's Disease and Parkinson's Disease: The Role of the Antioxidant Melatonin

Jana Tchekalarova et al.

Summary: Aging and neurodegenerative diseases, such as Alzheimer's disease and Parkinson's disease, are linked by common features like mitochondrial dysfunction and protein aggregation. The rising costs of caring for elderly patients with these diseases is a major issue in today's demographic crisis. Oxidative stress and imbalanced antioxidant defense system play crucial roles in age-related neurodegeneration, and diminished melatonin release with aging further increases susceptibility to these diseases. Understanding the underlying mechanisms and melatonin's role is important for developing precise treatment strategies.

Inhibiting nighttime melatonin and boosting cortisol increase patrolling monocytes, phagocytosis, and myelination in a murine model of multiple sclerosis

Majid Ghareghani et al.

Summary: The study found that melatonin functions through its receptors in the brain of multiple sclerosis (MS) patients. Excessive intake of melatonin exacerbates inflammation and immune cell migration to lesion areas. Therefore, monitoring the intake of melatonin in patients is necessary during treatment.
