SpliceVault predicts the precise nature of variant-associated mis-splicing

Summary: Even for splice-site variants that are likely to alter mRNA splicing, accurately predicting the resulting mis-splicing events is challenging. However, analyzing unannotated splicing events in large reference RNA-sequencing samples can predict the nature of variant-associated mis-splicing with high sensitivity. This method outperforms other predictive methods and is valuable for clinical interpretation and classification of splice-site variants.

Drug Discovery Perspectives of Antisense Oligonucleotides

Summary: The era of innovative RNA therapies using ASOs, siRNAs, and mRNAs is beginning. ASOs, targeting rare genetic diseases, have been developed into drugs and accepted as a powerful modality for next-generation medicines. They can theoretically target all disease-related RNAs, including protein-coding RNAs and non-coding RNAs, and regulate gene expression through different mechanisms. This review summarizes the achievements in medicinal chemistry, molecular mechanisms of action, structure-activity relationship, pharmacology, pharmacokinetics, and toxicology of ASOs, as well as recent advances in improving their therapeutic potential.

Drugging p53 in cancer: one protein, many targets

However, recent years have seen promising developments in p53-based therapy, including improved strategies and novel approaches. Small molecules that protect or restore p53 function are gaining interest, along with tailored drugs for specific p53 mutants. Additionally, gene therapy and immunotherapy are being reconsidered. However, significant challenges remain.

Systematic identification of intron retention associated variants from massive publicly available transcriptome sequencing data

Nonsense-mediated RNA decay: an emerging modulator of malignancy

Summary: Nonsense-mediated RNA decay (NMD) is a highly conserved RNA turnover pathway that selectively degrades RNAs harboring truncating mutations, which can affect the expression of tumor suppressor genes and oncogenes. NMD may have a dual role in tumors, as it can promote tumor growth by downregulating the expression of tumor suppressor genes and degrade immunogenic neoantigen peptides, but it can also suppress tumor generation and progression by degrading specific normal mRNAs and non-coding RNAs beneficial for tumors. NMD-modulatory therapy has the potential to provide widespread therapeutic benefit against diverse tumor types, but careful analysis is needed to determine whether NMD should be stimulated or repressed in specific tumors.

Drugging undruggable genes for cancer treatment: Are we making progress?
