Disposable, pressure-driven, and self-contained cartridge with pre-stored reagents for automated nucleic acid extraction

Summary: In this study, a pressure-driven and self-contained cartridge with pre-stored reagents was designed to automate nucleic acid purification. By using pressurized air stored inside the cartridge, reagents were transferred through a microfluidic chip, which reduced operational complexity. The device achieved similar results to the conventional column-based extraction method in terms of concentration, purity, and real-time polymerase chain reaction (qPCR) for Escherichia coli (E. coli) O157:H7 DNA. It also had a detection limit of 103 CFU, similar to the conventional solid-phase extraction method. This novel approach for automating sample preparation can facilitate point-of-care molecular diagnostics.

A high quality, high molecular weight DNA extraction method for PacBio HiFi genome sequencing of recalcitrant plants

Summary: This study developed a DNA extraction method for long-read sequencing of recalcitrant plants. By modifying the lysis buffer, eliminating traditional purification steps, and optimizing cell/nucleus lysis, high quality, high molecular weight DNA suitable for PacBio HiFi sequencing was successfully extracted. The obtained sequences showed high contiguity and provided a good draft assembly. The developed DNA extraction method is compatible with PacBio HiFi sequencing and suitable for de novo whole genome sequencing projects of plants.

However, the overgrowth of Agrobacterium is a major concern that hampers the transformation process. This phenomenon can spoil the entire transformation process and is difficult to cure once it occurs. The regulation of various factors, such as explant nature, A. tumefaciens strain, T-DNA vector, co-cultivation conditions, acetosyringone, washing medium, and antibiotics, can help prevent this phenomenon and control the overgrowth of Agrobacterium.

The DNA microlibrary@MOFs enhances the stability of data-encoded DNA against harsh environments and can be read out perfectly after accelerated aging, equivalent to 10-year storage under specific conditions.

Fast and visual detection of nucleic acids using a one-step RPA-CRISPR detection (ORCD) system unrestricted by the PAM

Summary: We developed a one-step RPA-CRISPR detection system (ORCD) that can detect nucleic acids with high sensitivity and specificity without restriction of the PAM sequence. In this system, Cas12a detection and RPA amplification are performed simultaneously, and 0.2 copies/μL of DNA and 0.4 copies/μL of RNA can be detected. By combining this detection technique with a nucleic acid extraction-free method, our ORCD system can extract, amplify, and detect samples within 30 minutes, and shows high sensitivity and specificity compared to PCR.

However, concerns about their adverse health and environmental effects, as well as evidence of transgene leakage to crops, have raised questions about their benefits and risks. This study developed a closed tube colorimetric GMO detection method based on Loop-Mediated Isothermal Amplification (LAMP) technique, which can be integrated into GMO-detection platforms. The durability of the LAMP mixture in test tubes showed that it could yield DNA amplicons for up to 3 days at room temperature and 6 days at 4 degrees Celsius.
