Deep Crypt Secretory Cell Differentiation in the Colonic Epithelium Is Regulated by Sprouty2 and Interleukin 13

Summary: This study found that Sprouty2 limits the differentiation of colonic deep crypt secretory (DCS) cells by suppressing the IL13 signaling pathway. The DCS cells express high levels of the antimicrobial peptide RELM beta in homeostasis. Loss of Sprouty2 leads to the release of IL13, promoting the proliferation of DCS cells and increased levels of colonic RELM beta.

Summary: TFF2 is a protein that is co-secreted with MUC6 in the gastric mucous neck cells, antral gland cells, and duodenal Brunner glands. It forms a high-molecular-mass complex with MUC6, stabilizing the mucus barrier. In the pancreas, TFF2 exists as a low-molecular-mass monomer. Its absence in the pancreas is due to a lack of MUC6. Tff2 plays a protective role in the pancreatic ductal epithelium.

Profiling of the Bacterial Microbiota along the Murine Alimentary Tract

The high-molecular mass form is TFF3-FCGBP oligomers, while the low-molecular mass forms are homodimeric and monomeric TFF3 with potentially anti-apoptotic activities. Disulfide-linked TFF3 heterodimers were also detected in bronchial secretions and BAL fluid. The study discusses a hypothetical model for how TFF3 may control FCGBP oligomerization and found no interactions between TFF3-FCGBP and DMBT1gp(340) or MUC5AC.

Summary: Intestinal goblet cells are not homogeneous, with several distinct subtypes forming two differentiation trajectories. Intercrypt goblet cells located at the colonic luminal surface produce mucus with different properties from crypt-residing goblet cells. Defective icGCs in mice lead to increased sensitivity to chemically induced colitis and spontaneous colitis with age, while alterations in mucus and reduced numbers of icGCs are observed in patients with ulcerative colitis, highlighting the importance of icGCs in maintaining functional epithelial protection.
