Mitochondrial membrane proteins and VPS35 orchestrate selective removal of mtDNA

Summary: This study reveals the mechanisms underlying excessive turnover of mtDNA following specific mtDNA damage, which relies on lysosomal function and ATG5 rather than canonical macroautophagy. The study also demonstrates that the targeting of mitochondrial nucleoids is controlled by the ATAD3-SAMM50 axis and that mtDNA is eliminated through endosomes mediated by VPS35. These findings have important implications for understanding mtDNA-related diseases and improving mitochondrial dysfunction.

Short-lived proteins are less abundant, evolutionarily younger, and less thermally stable compared to other proteins, with 103 proteins showing different stabilities among cell lines. Truncated forms of certain proteins in U2OS and HCT116 cells were also identified, indicating lower stability than full-length counterparts. This research provides a large-scale resource for understanding human short-lived proteins and their potential for therapeutic interventions.

Summary: The acidic activation domain (AD) of the yeast transcription factor Gal4 acts through binding to the Med15 subunit of the Mediator complex. The interaction between Gal4 and Med15 occurs through the same fuzzy binding mechanism as Gcn4 AD, indicating a common sequence-independent mechanism for AD-Mediator binding. Conversely, the structured binding partner Gal80 determines the type of protein-protein interaction for the intrinsically disordered protein Gal4 AD.
