Influence of ultrasonication and hydrolysis conditions in methylation analysis of bacterial homoexopolysaccharides

Summary: Homoexopolysaccharides (HoEPS) like alpha-glucans and beta-fructans are synthesized by certain types of bacteria. Methylation analysis is an important tool for studying the structure of these polysaccharides, but the conditions for analysis need to be adjusted for different types of HoEPS. Ultrasonication is crucial for swelling and deprotonation of water insoluble alpha-glucans, while water soluble HoEPS do not require ultrasonication. Acid hydrolysis conditions also differ for different HoEPS. Overall, our results demonstrate the need for customized conditions in methylation analysis of bacterial HoEPS.

Summary: High molecular weight dextrans from different bacterial strains exhibit varying effects on dextran production, metabolic fluxes, lactate and mannitol synthesis. One strain showed increased binding to cells and plates under conditions for dextran synthesis, demonstrating a positive influence of dextran on adhesion properties.

Summary: Exopolysaccharides (EPS) from Streptococcus thermophilus offer similar technofunctionality as commercial thickeners at lower concentrations, but are not widely used in non-fermented foods. The purity, molecular weight, and intrinsic viscosity of EPS do not have direct correlation, and the addition of EPS can increase the gel stiffness of products.

Summary: This review highlights the advantages of using dextran in breadmaking, including increasing bread volume, softness and moisture content, as well as retarding starch retrogradation. Dextran with high molecular weight and less branching has a superior effect on enhancing bread quality, while mild acidification favors the functionality of dextran in dough and bread systems.
