DGKB mediates radioresistance by regulating DGAT1-dependent lipotoxicity in glioblastoma

Summary: Glioblastoma (GBM) cells that survive radiotherapy contribute to tumor progression and recurrence with metabolic advantages. The downregulation of diacylglycerol kinase B (DGKB) and upregulation of diacylglycerol acyl-transferase 1 (DGAT1) confer radioresistance by reducing mitochondrial lipotoxicity. Cladribine, a clinical drug, activates DGKB, inhibits DGAT1, and sensitizes GBM cells to radiotherapy.

Radioresistance and Transcriptional Reprograming of Invasive Glioblastoma Cells

Emerging therapies for glioblastoma: current state and future directions

Immunotherapy, including immune checkpoint blockade, CAR T cell therapy, oncolytic virotherapy, and vaccine therapy, has shown promising results in improving GBM outcomes. Techniques to overcome the blood-brain barrier for targeted delivery are also being tested. This article reviews the rationales for these therapies, potential novel agents, current status of trials, and discusses challenges and future perspectives in glioblastoma immuno-oncology.

Glioblastoma hijacks neuronal mechanisms for brain invasion

Summary: A subpopulation of glioblastoma cells lacking connections with other tumor cells and astrocytes but receiving synaptic input from neurons drives whole-brain colonization. These cells resemble neuronal and neural-progenitor-like states and invade the brain using mechanisms similar to neuronal migration. Neuronal activity induces calcium signals, leading to the formation of tumor microtubes and increased invasion speed. This study reveals how neuronal mechanisms govern glioblastoma cell invasion and explains the close interlink between tumor dissemination and cellular heterogeneity.

Cellular sensitivity to radiation is correlated with the ability to repair DNA lesions, and recent studies have used whole-genome analysis to identify genes that impact individual sensitivity.

Discovery of DRP-104, a tumor-targeted metabolic inhibitor prodrug

Summary: 6-Diazo-5-oxo-l-norleucine (DON) is a glutamine antagonist that suppresses cancer cell metabolism while enhancing the metabolic fitness of tumor CD8+ T cells. The development of DON was halted due to dose-limiting gastrointestinal toxicities. In order to overcome this, a DON peptide prodrug, DRP-104, was designed and showed improved tolerability and enhanced efficacy in tumor regression. The effect of DRP-104 is dependent on CD8+ T cells and results in strong immunologic memory.

This deficiency affects the malate-aspartate shuttle, gluconeogenesis, amino acid homeostasis, and the urea cycle.
