Microdialysis of Drug and Drug Metabolite: a Comprehensive In Vitro Analysis for Voriconazole and Voriconazole N-oxide

Josefine Schulz et al.

Summary: The feasibility of simultaneous microdialysis of voriconazole (VRC) and its metabolite N-oxide (NO) was explored in this in vitro study. The relative recovery (RR) of both compounds was investigated and quantified, and it was found that there were no significant differences in RR between individual and simultaneous microdialysis. RR was not dependent on concentration. The study day was the main source of variability, while the microdialysis catheter had a minor effect. These results support the application of microdialysis in clinical trials to assess target-site concentrations of VRC and NO.

Quantification of microdialysis related variability in humans: Clinical trial design recommendations

David Busse et al.

Summary: This study investigated and analyzed the variability of RR values in non-obese and obese patients, finding that patient-related factors were the main contributor to variability in RR, followed by factors related to the catheter. Fat mass in obese patients affected RR, but only explained a portion of the variability.

Microdialysis sampling to monitor target-site vancomycin concentrations in septic infants: a feasible way to close the knowledge gap

Sebastian Schroepf et al.

Summary: This study, dedicated to the memory of pharmacokinetics pioneer Hartmut Derendorf, explores the feasibility and safety of microdialysis sampling for vancomycin monitoring in septic infants/neonates. The results demonstrated the applicability of microdialysis for vancomycin monitoring, with well-tolerated sampling and plausible pharmacokinetic profiles obtained. Further larger trials are warranted to characterize the pharmacokinetics and variability of vancomycin at the target site in vulnerable patients.

Evaluation of Benzylpenicillin as an Internal Standard for Measurement of Piperacillin Bone Concentrations Via Microdialysis

Martin Knudsen et al.

Summary: The study indicates that benzylpenicillin is suitable as an internal standard for piperacillin in microdialysis studies. The mean fT>MIC (16 mg/mL) reached a target of 50% fT>MIC, but not all pigs in all compartments achieved this target. Different strategies must be considered for higher targets to be met.

A sensitive and high-throughput quantitative liquid chromatography high-resolution mass spectrometry method for therapeutic drug monitoring of 10 β-lactam antibiotics, linezolid and two β-lactamase inhibitors in human plasma

Sarah Van Vooren et al.

Summary: An UHPLC-HRMS method was developed for simultaneous and sensitive quantification of 10 beta-lactam antibiotics, linezolid, and beta-lactamase inhibitors in human plasma. The method showed high sensitivity and accuracy, allowing it to be suitable for routine therapeutic drug monitoring.

Piperacillin-Tazobactam in Intensive Care Units: A Review of Population Pharmacokinetic Analyses

Ibrahim El-Haffaf et al.

Summary: This review summarizes population pharmacokinetic models for Piperacillin-tazobactam in the intensive care unit, providing comprehensive data on dosing strategies and identifying significant covariates in critically ill patients. Studies showed variability in drug clearance and volume of distribution, with creatinine clearance and body weight being common covariates. Continuous or extended infusion methods were found to be more effective than intermittent administration in achieving appropriate pharmacodynamic targets.
