AP-1 activation mediates post-natal cardiomyocyte maturation

Summary: Our study shows that activating protein 1 (AP-1) transcription factors play a crucial role in post-natal cardiomyocyte maturation. Activating members of the AP-1 family inhibits cardiomyocyte cytokinesis and promotes maturation, while knocking down AP-1 members promotes cytokinesis and inhibits maturation. Mechanistically, AP-1 members activate the expression of fatty acid metabolic genes to promote cardiomyocyte maturation. Furthermore, inhibition of AP-1 using T-5224 also inhibits cardiomyocyte maturation in neonatal mice.

A CCR4 antagonist attenuates atopic dermatitis-like skin inflammation by inhibiting the recruitment and expansion of Th2 cells and Th17 cells

Summary: CCR4 is a major trafficking receptor for T-helper (Th) 2 cells and Th17 cells and is considered as a potential therapeutic target for atopic dermatitis (AD). CCR4 antagonists may exhibit anti-allergic effects by inhibiting both the recruitment and expansion of Th2 cells and Th17 cells in AD. This study demonstrated that the CCR4 inhibitor, compound 22, ameliorated AD-like skin lesions by reducing the number of Th2 cells and Th17 cells in the skin lesions and regional lymph nodes (LNs).
