Soluble TREM2 in CSF and its association with other biomarkers and cognition in autosomal-dominant Alzheimer's disease: a longitudinal observational study

Estrella Morenas-Rodriguez et al.

Summary: This study aimed to investigate the associations between soluble TREM2, a biomarker of TREM2 signaling, and amyloid beta deposition, tau-related pathology, neuroimaging markers, and cognitive decline in the progression of Alzheimer's disease. The results demonstrated that increased soluble TREM2 was associated with amyloid beta deposition, tau pathology, cortical shrinkage, and cognitive decline.

Elevating microglia TREM2 reduces amyloid seeding and suppresses disease-associated microglia

Na Zhao et al.

Summary: This study using mouse models demonstrates that overexpression of human TREM2 in microglia can reduce amyloid deposition and suppress disease-associated microglia in the early amyloid seeding stage, while the R47H variant exacerbates amyloid burden in the middle amyloid rapid growth stage. These findings highlight the dynamic effects of TREM2 in modulating AD pathogenesis and emphasize the beneficial effect of enhancing TREM2 function in the early stage of AD development.

Summary: The study showed that sTREM2 fragments 41-81 and 51-81 enhanced cell viability and inflammatory responses in primary microglia; fragment 41-81 was more efficient than 51-81 in ameliorating amyloid-related pathology and is a promising candidate for AD therapy.

TREM2/PLCγ2 signalling in immune cells: function, structural insight, and potential therapeutic modulation

Lorenza Magno et al.

Summary: The central role of resident innate immune cells (microglia) in neurodegeneration has been highlighted through recent studies, particularly focusing on the gene products TREM2 and PLC gamma 2. These insights offer new opportunities for the development of novel therapies in neurodegenerative disorders.
