We have reached a dead end for preimplantation genetic testing for aneuploidy

Norbert Gleicher et al.

Summary: The hypothesis of preimplantation genetic testing for aneuploidy (PGT-A) was proposed 20 years ago, but recent studies have shown that its application does not have significant advantages and may even result in adverse outcomes when patient selection biases are reversed. Additionally, high embryo mosaicism leads to the unnecessary discarding of normal embryos, causing harm to patients. Therefore, it is recommended to restrict PGT-A to research protocols and to encourage professional societies to issue appropriate practice guidelines.

IVF outcomes of embryos with abnormal PGT-A biopsy previously refused transfer: a prospective cohort study

D. H. Barad et al.

Summary: The study found that many patients choose to transfer embryos diagnosed as abnormal to a new institution for transfer, recognizing that these embryos can still offer a chance of pregnancy and live birth. While not all abnormal embryos were successfully transferred, some patients were able to achieve live births through this method.

PGDIS position statement on the transfer of mosaic embryos 2021

D. Leigh et al.

Summary: Chromosome testing strategies, such as PGT-A, improve initial IVF outcomes by avoiding transfer of aneuploid embryos. Newer technologies have revealed trophectoderm mosaicism in embryos with intermediate copy number results. The transfer of mosaic embryos may be associated with increased implantation failure and miscarriage rates.

Single-cell multiomics analyses of spindle-transferred human embryos suggest a mostly normal embryonic development

Shuyue Qi et al.

Summary: Spindle transfer (ST) is a potential method to prevent the transmission of mitochondrial DNA mutations. This study used single-cell triple omics sequencing to evaluate the safety of ST and found that it is generally safe for embryonic development, with only a minor delay in the DNA demethylation process at the blastocyst stage.

Regional and developmental characteristics of human embryo mosaicism revealed by single cell sequencing

Yixin Ren et al.

Summary: The study revealed that around 60-65% of blastocysts were mosaic at single-cell sequencing level in TE and ICM cells, while the corresponding hESCs showed a significantly lower rate of mosaicism compared to TE and ICM cells. This suggests a potential decline in mosaicism incidence during embryonic development.

Limits imposed by the experimental design of a large prospective non-inferiority study on PGT-A invalidate many of the conclusions

Summary: The experimental design and data interpretation in the study had serious flaws, leading to the inability to support the major conclusions presented. Preimplantation genetic testing for aneuploidy (PGT-A) was underutilized, resulting in no potential for enhanced cumulative outcomes.

The Multifaceted Regulation of Mitochondrial Dynamics During Mitosis

Evanthia Pangou et al.

Summary: Mitosis ensures genome integrity by precisely segregating genetic material, while also tightly coordinating the segregation of subcellular organelles, such as mitochondria, to meet energy demands and facilitate their efficient inheritance. Mitochondria, highly dynamic organelles, undergo fission, fusion, and transport events during the cell cycle, with kinases phosphorylating key factors to drive fragmentation for distribution to daughter cells. This review highlights the complex regulatory networks linking mitotic factors, mitochondrial dynamics, and potential implications in human disease.
