Novel TARS2 variant identified in a Chinese patient with mitochondrial encephalomyopathy and a systematic review

Peiqing He et al.

Summary: Biallelic pathogenic variants in the TARS2 gene cause COXPD21, a rare mitochondrial encephalomyopathy characterized by early-onset hypotonia, developmental delay, seizures, and brain anomalies. This study reports a Chinese female with COXPD21 and identifies novel mutations in the TARS2 gene, expanding the understanding of the genotype-phenotype relationship in COXPD21.

Refining the mutational spectrum and gene-phenotype correlates in pontocerebellar hypoplasia: results of a multicentric study

Sara Nuovo et al.

Summary: Pontocerebellar hypoplasias (PCH) are a group of genetically heterogeneous disorders, with CASK being the major causative gene in Italy. The most common genetic causes include CASK, TSEN54, and EXOSC3, with a clear association observed between CASK and specific phenotypic clusters. Variability in phenotypes associated with the most common genetic causes of PCH is wider than previously thought, with marked overlap between CASK and TSEN54-associated disorders.

Elucidating the molecular mechanisms associated with TARS2-related mitochondrial disease

Wen-Qiang Zheng et al.

Summary: TARS2 gene encodes a mitochondrial enzyme crucial for mitochondrial protein synthesis, and its pathogenic variants can lead to mitochondrial diseases. New cases reported in this study have expanded the clinical spectrum of TARS2-related mitochondrial diseases, demonstrating the pathogenic nature of these variants.

tRNA dysregulation and disease

Esteban A. Orellana et al.

Summary: tRNA function and gene expression are dynamically regulated by post-transcriptional modifications. tRNAs play an important role in protein synthesis and show tissue-specific and cell type-specific expression patterns. Dysregulation of tRNAs can contribute to various human diseases, and controlling functional tRNA levels is crucial for mRNA translation and protein expression.

Mitochondrial Threonyl-tRNA Synthetase TARS2 Is Required for Threonine-Sensitive mTORC1 Activation

Sung-Hoon Kim et al.

Summary: This study reveals a new pathway for activating mTORC1 in response to variations in threonine levels through mitochondrial synthetase TARS2. The interaction between TARS2 and Rags increases GTP loading of RagA, influencing the activity of mTORC1.

Phenotypic diversity of brain MRI patterns in mitochondrial aminoacyl-tRNA synthetase mutations

Charles-Joris Roux et al.

Summary: Mitochondrial aminoacyl-tRNA synthetases encoded by ARS2 genes play a critical role in mitochondrial translation accuracy, with mutations linked to various clinical presentations affecting the CNS. This study analyzed brain MRI scans of 25 patients with biallelic pathogenic variants of 11 ARS2 genes, identifying specific MRI patterns associated with certain mutations while also highlighting cases where MRI results appeared normal despite the presence of ARS2 mutations, suggesting potential underdiagnosis of ARS2 mutations.

Biallelic variants in KARS1 are associated with neurodevelopmental disorders and hearing loss recapitulated by the knockout zebrafish

Sheng-Jia Lin et al.

Summary: Pathogenic variants in Lysyl-tRNA synthetase 1 (KARS1) have been increasingly recognized as a cause of early-onset complex neurological phenotypes. Through international collaboration, 22 affected individuals from 16 unrelated families with biallelic likely pathogenic or pathogenic KARS1 variants were identified. Loss-of-function alleles in zebrafish were generated to mimic key disease phenotypes. The study delineates the clinical spectrum associated with KARS1 defects and suggests potential therapeutic targets related to p53 signaling components.
