Synthesis, antioxidant and anti-diabetic potential of novel benzimidazole substituted coumarin-3-carboxamides

Summary: A series of benzimidazole tethered coumarin-3-carboxamide analogues were synthesized and tested for their antioxidant potential using DPPH and ABTS assays. The compounds also demonstrated moderate alpha-amylase inhibition. Compound 10j showed the highest DPPH radical scavenging activity with an IC50 value of 89.57 μM, while compound 10f exhibited the strongest scavenging activity against ABTS radicals with an IC50 value of 93.45 μM. Compound 10f also emerged as the best antidiabetic analogue with an IC50 value of 67.52 μM.

New Succinimide-Thiazolidinedione Hybrids as Multitarget Antidiabetic Agents: Design, Synthesis, Bioevaluation, and Molecular Modelling Studies

Summary: Diabetes mellitus (DM) is a metabolic disorder resulting from pancreatic dysfunction, characterized by reduced insulin production or insulin resistance. In this study, we designed and synthesized new succinimide-thiazolidinedione hybrids for the treatment of DM. We successfully synthesized five new derivatives (10a-e) with different substitution patterns on the N-atom of the succinimide ring. Compound 10d showed the most potential as an antidiabetic agent in in vitro assays. Further investigations in experimental animals confirmed its promising antidiabetic activity. Molecular docking studies also demonstrated the interaction of compound 10d with target enzymes involved in diabetes. Overall, we have identified a new class of compounds with potential as antidiabetic agents.

Summary: This study designed a new compound that can modulate multiple targets related to hyperglycemia simultaneously. The compound exhibited excellent inhibition against DPP-4 activity and good antioxidant potential in vitro, as well as safety and effectiveness in vivo experiments. Docking studies were also conducted to investigate the binding orientations and interactions with key amino acid residues of the compound and the selected targets.

Summary: The synthesis of novel 1,3-thiazoles and 4-thiazolidinones demonstrated promising activity against Leishmania spp. and Trypanosoma cruzi, with compound 1f showing the most potent effect. Additionally, compound 1f induced necrosis and apoptosis in T. cruzi, leading to parasite death. This study provides potential avenues for scaffold optimization in the development of improved and safer drug candidates for neglected tropical diseases.
