Hybrids of thiazolidinone with 1,2,3-triazole derivatives: design, synthesis, biological evaluation, in silico studies, molecular docking, molecular dynamics simulations, and ADMET profiling

Abdoullah Bimoussa et al.

Summary: A series of thiazolidinone linked 1,2,3-triazole hybrids were synthesized using the CuAAC reaction, and their cytotoxic activity against human cancer cell lines was evaluated. Compound 5a showed the highest activity against HT-1080 and MCF-7 cell lines. Docking studies and molecular dynamic simulations confirmed the experimental results. The derivatives 5a and 5d exhibited comparable docking scores, and 5a showed higher stability in complex formation with protein p53. Both derivatives showed good drug-like properties and safety.

Diastereomerically pure (S)-verbenone-isoxazoline hybrids: Synthesis, molecular structure, Hirshfeld surface analysis, density functional theory, cytotoxic effect, and apoptosis induction

Mourad Fawzi et al.

Summary: In this study, a new diastereomerically pure isoxazoline compound was efficiently synthesized from (S)-verbenone. The structure of the compound was confirmed using various analytical and spectral techniques, and its properties and activities were investigated. Compound 3d exhibited significant cytotoxic activity against HT-1080 and A-549 cancer cells, inducing apoptosis and cell cycle arrest.

Novel isoxazoline-linked 1,3,4-thiadiazole hybrids as anticancer agents: Design, synthesis, biological evaluation, molecular docking, and molecular dynamics simulation

Ali Oubella et al.

Summary: In this study, isoxazoline derivatives with 1,3,4-thiadiazole moiety were efficiently synthesized using natural (R)-carvone as a starting material. The synthesized compounds exhibited moderate to high anticancer activity and could induce apoptosis through caspase-3/7 activation.

Novel Hydrazono-2-Iminothiazolidin-4-Ones Based on a Monoterpenic Skeleton as Potential Antitumor Agents: Synthesis, DFT Studies, in Vitro Cytotoxicity, Apoptosis Inducing Properties and Molecular Docking

Mourad Fawzi et al.

Summary: A series of novel compounds were synthesized from limonaketone and evaluated for their cytotoxic activity against human cancer cell lines. Thiazolidinones 9 and 10 showed strong anticancer activity in HT-1080 cell lines, with compound 10 exhibiting the highest ability to induce apoptosis and activate caspase-3/7. Molecular docking analysis indicated that compounds 9 and 10 formed stable ligand-caspase-3 complexes, with the presence of a phenyl group in compound 10 enhancing caspase-3 activation through additional hydrogen bonds with specific amino acids.

Therapeutic progress and challenges for triple negative breast cancer: targeted therapy and immunotherapy

Ruoning Yang et al.

Summary: Triple negative breast cancer (TNBC) is a subtype of breast cancer that lacks estrogen receptor, human epidermal growth factor receptor 2, and progesterone receptor. Current treatment options are limited and primarily involve surgery and chemotherapy, which show limited effectiveness in advanced stages. The lack of therapeutic targets and hormone receptors in TNBC contributes to the limited treatment options, thus highlighting the need for more effective treatments and predictive biomarkers.

Synthesis, structure and in vitro antiproliferative effects of alkyne-linked 1,2,4-thiadiazole hybrids including erlotinib- and ferrocene-containing derivatives

Mohammed Boulhaoua et al.

Summary: Chemotherapy is crucial in cancer treatment, and developing new drugs with minimal side effects is important. A series of novel 1,2,4-thiadiazoles bearing erlotinib were synthesized and tested for anti-proliferative effects against various cancer cell lines. Some compounds showed promising anti-cancer activity, particularly the isomer mixture of bis-erlotinib substituted thiadiazoles.
