Down-regulation miR-146a-5p in Schwann cell-derived exosomes induced macrophage M1 polarization by impairing the inhibition on TRAF6/NF-?B pathway after peripheral nerve injury

Jun Sun et al.

Summary: This study focused on investigating the roles and mechanisms of exosomes derived from Schwann cells under different conditions in regulating macrophagic sub-phenotypes and peripheral nerve injury repair. The study found that exosomes from normal oxygen condition Schwann cells promoted M2 macrophagic polarization and facilitated axon elongation, while exosomes from post-injury oxygen-glucose-deprivation-condition Schwann cells promoted M1 polarization and inhibited axon elongation. The down-regulation of miR-146a-5p was closely related to the shift of exosomes to pro-inflammatory phenotype. The study concluded that miR-146a-5p played an important role in regulating macrophagic phenotype and had implications for axonal regeneration and functional recovery.

PGAM5 regulates DRP1-mediated mitochondrial fission/mitophagy flux in lipid overload-induced renal tubular epithelial cell necroptosis

Dan Sang et al.

Summary: This study investigates the mechanism of PGAM5 in promoting lipid-induced necroptosis in renal tubular epithelial cells (RTECs). The study finds that upregulation of PGAM5 activates the necroptosis pathway, leading to cell death. However, silencing PGAM5 can prevent these changes, protecting RTECs from lipid-induced necroptosis.

Schwann cell-derived exosomes containing MFG-E8 modify macrophage/microglial polarization for attenuating inflammation via the SOCS3/STAT3 pathway after spinal cord injury

Jie Ren et al.

Summary: The study demonstrates that Schwann cell-derived exosomes (SCDEs) can attenuate inflammation in spinal cord injury (SCI) by suppressing M1 polarization and stimulating M2 polarization. SCDEs also improve the recovery of SCI in rats and MFG-E8 is identified as the main component that improves the inflammatory response. The findings provide important insights for the clinical translation of SCDEs treatment for SCI.

The role of biomechanical stress in extracellular vesicle formation, composition and activity

Will Thompson et al.

Summary: Extracellular vesicles (EVs) play a crucial role in intercellular communication and have important applications in basic research, clinical medicine, and biotechnology. However, the variability in EV composition caused by culture conditions poses challenges in bioprocessing. Understanding the relationship between EV production and composition is becoming increasingly important. While chemical stimulation and culture conditions are known factors that affect EV biology, the impact of biomechanical forces on EV generation, properties, and biological activity is not well understood. This review provides a comprehensive evaluation of the influence of biomechanical stresses on EV production, composition, and biological activity, and discusses potential implications for scalable bioprocessing and quality control.

Mitochondrial dysfunction as a target in spinal cord injury: intimate correlation between pathological processes and therapeutic approaches

Julieta Schmidt et al.

Summary: Traumatic spinal cord injuries cause disruption of axonal projections, resulting in impaired locomotor, sensory, and autonomic functions. Mitochondrial dysfunction in the injured spinal tissue prevents the regeneration of severed axons, leading to sustained impairment in voluntary control of movement. This review discusses the link between mitochondrial dysfunction and axonal regeneration failure in spinal cord injury, and explores potential mitochondria-targeted therapeutic approaches to promote functional recovery.

Spinal cord injury: molecular mechanisms and therapeutic interventions

Xiao Hu et al.

Summary: Spinal cord injury (SCI) is a severe condition with a high disability rate. The challenges in SCI repair lie in its complex pathological mechanisms and the difficulty of neural regeneration in the central nervous system. Recent advancements in single-cell sequencing and spatial transcriptome analysis have revealed new pathological mechanisms of SCI, particularly the interactions between immune and neural cell responses. Stem cell therapy, biomaterial transplantation, and electromagnetic stimulation have entered clinical trials, showing promise for future SCI repair and clinical applications.
