Number of galloyl moiety and intramolecular bonds in galloyl-based polyphenols affect their interaction with alpha-glucosidase

Summary: The study found that the inhibitory activity of alpha-glucosidase increased with the number of free galloyl moieties, while unfree galloyl moieties mainly contributed to inhibition through ellagic acid and hexahydroxydiphenoyl group. The polyphenols could form a static complex with the enzyme through binding interactions, and the number of amino acid residues involved in these interactions was positively correlated with inhibitory activity.

Inhibitory Mechanism of Baicalein on Acetylcholinesterase: Inhibitory Interaction, Conformational Change, and Computational Simulation

Summary: In this study, baicalein, a bioactive flavonoid, was found to competitively inhibit acetylcholinesterase (AChE) through a static process. The binding of baicalein to AChE was mainly driven by hydrogen bonding and hydrophobic interaction, resulting in structural changes of AChE. Molecular docking and dynamics simulations confirmed the inhibitory effect of baicalein on AChE. This study provides insights into the molecular-level understanding of the interaction between baicalein and AChE, which may contribute to the development of anti-Alzheimer's disease functional foods and drugs.

Screening and characterization of potential α-glucosidase inhibitors from Cercis chinensis Bunge fruits using ultrafiltration coupled with HPLC-ESI-MS/MS

Summary: A more accurate HPLC-MS screening method was developed by combining functional enzyme assay and affinity ultrafiltration screening assay for the screening of natural product inhibitors of alpha-glucosidase from Cercis chinensis Bunge fruits. Twelve bioactive components targeting alpha-glucosidase were identified using this method, with known inhibitor acarbose used as a positive control to eliminate false positives. Molecular docking confirmed the results of the screening assay by investigating the bindings of the twelve components to the active site of alpha-glucosidase.

Screening and identification of a-glucosidase inhibitors from Cyclocarya paliurus leaves by ultrafiltration coupled with liquid chromatography-mass spectrometry and molecular docking

Summary: The study developed an affinity-based screening assay to identify potential alpha-glucosidase inhibitors from Cyclocarya paliurus leaves. A total of 36 active compounds were screened and further characterized by UPLC-QTOF-MS/MS, with molecular docking revealing possible inhibitory mechanisms between active compounds and alpha-glucosidase. The results suggest that C. paliurus has a promising potential as a candidate for the prevention of diabetes mellitus.
