Role of Mitochondrial Cytochrome P450 2E1 in Healthy and Diseased Liver

Julie Massart et al.

Summary: CYP2E1 plays a pivotal role in hepatotoxicity induced by alcohol abuse and different xenobiotics, generating reactive metabolites that induce oxidative stress, mitochondrial dysfunction, and cell death. Increased CYP2E1 activity in nonalcoholic fatty liver disease (NAFLD) leads to reactive oxygen species overproduction, triggering oxidative stress, necroinflammation, and fibrosis. Mitochondrial CYP2E1 (mtCYP2E1) has been found in different experimental models and may contribute to alcohol-associated liver disease, xenobiotic-induced hepatotoxicity, and NAFLD.

Insights into the role of STAT3 in intrahepatic cholangiocarcinoma

Ranzhiqiang Yang et al.

Summary: Intrahepatic cholangiocarcinoma (ICC) is a primary malignant liver tumor with a poor prognosis, characterized by neoplastic transformation of intrahepatic biliary epithelial cells. Signal transducer and activator of transcription 3 (STAT3) is an important oncogene that may serve as a target for ICC treatment. Further research is needed to improve understanding of STAT3 and its role in ICC development.

Hypoxia activated HGF expression in pancreatic stellate cells confers resistance of pancreatic cancer cells to EGFR inhibition

Xiuhui Shi et al.

Summary: High expression levels of alpha-SMA and hypoxia markers are associated with poor prognosis in pancreatic cancer patients. Mechanistically, hypoxia induces HGF expression and secretion in PSC via HIF-1 alpha, which then activates Met and suppresses pancreatic cancer cell sensitivity to EGFR inhibitors. The combination of EGFR inhibitor and Met inhibitor shows promise for pancreatic cancer treatment based on the demonstrated signaling axis between PSC and cancer cells.

Hepatic inactivation of murine Surf4 results in marked reduction in plasma cholesterol

Vi T. Tang et al.

Summary: This study reveals the importance of SURF4 in the hepatic secretion of PCSK9 and APOB-containing lipoproteins. By generating mice with hepatic SURF4 deficiency, the researchers found that the absence of SURF4 leads to a severe defect in lipoprotein secretion, resulting in decreased plasma cholesterol and triglyceride levels. These findings highlight the potential of SURF4 as a therapeutic target in atherosclerotic cardiovascular diseases.

Ziziphus spina-christi leaf extract attenuates mercuric chloride-induced liver injury in male rats via inhibition of oxidative damage

Shimaa S. Ramadan et al.

Summary: Z. spina-christi leaf extract showed hepatoprotective effects against mercuric chloride-induced liver damage by reducing mercury concentration, oxidative stress, and inflammation while increasing antioxidant enzyme activity and activating the Nrf2/HO-1 signaling pathway. This extract also decreased expression of pro-inflammatory cytokines and pro-apoptotic proteins and increased expression of anti-apoptotic protein Bcl-2.

CYP2E1 in Alcoholic and Non-Alcoholic Liver Injury. Roles of ROS, Reactive Intermediates and Lipid Overload

Riina Harjumaki et al.

Summary: CYP2E1 is a highly conserved cytochrome P450 gene in the human genome, with important physiological functions including involvement in liver homeostasis, activation of procarcinogens, conversion of drugs to cytotoxic products, reduction of toxic compounds, and contribution to the development of alcoholic liver disease. Its fat-dependent induction may provide a link between steatosis and fibrosis in the liver.

Epigenomic and transcriptional profiling identifies impaired glyoxylate detoxification in NAFLD as a risk factor for hyperoxaluria

Kathrin Gianmoena et al.

Summary: Epigenetic modifications in NAFLD, particularly in glyoxylate metabolism, contribute to kidney damage via oxalate release. The downregulation and hypermethylation of AGXT in steatotic hepatocytes lead to elevated oxalate levels, explaining the increased risk of kidney stones and chronic kidney disease in NAFLD patients.

The Thioesterase ACOT1 as a Regulator of Lipid Metabolism in Type 2 Diabetes Detected in a Multi-Omics Study of Human Liver

Marco Cavalli et al.

Summary: The study integrated ATAC-seq data, multi-omics data, and CRE databases to identify candidate CREs associated with T2D, detecting differentially accessible regions in human liver samples from normal and T2D donors. A candidate enhancer for the ACOT1 gene was identified, linking its regulation to T2D. ACOTs are enzymes associated with regulation of triglyceride levels, fatty acid oxidation, mitochondrial function, and insulin signaling, with implications for T2D pathogenesis.

ACSS3 represses prostate cancer progression through downregulating lipid droplet-associated protein PLIN3

Lijie Zhou et al.

Summary: This study identified downregulation of ACSS3 in prostate cancer due to gene promoter methylation, with restoration of ACSS3 expression reducing LD deposits, promoting apoptosis, decreasing androgen synthesis, inhibiting CRPC progression, and reversing Enzalutamide resistance. Mechanistically, ACSS3 reduced LD deposits by regulating the stability of the LD coat protein PLIN3, thereby repressing prostate cancer progression.
