Improving thermostability of Bacillus amyloliquefaciens alpha-amylase by multipoint mutations

Summary: This study successfully improved the thermostability of medium-temperature alpha-amylase by altering certain amino acids in BAA. The analysis of predicted structures revealed that the mutant had more calcium-binding sites and intramolecular interactions, as well as reduced steric hindrance at the active cavity, which contributed to the increased thermostability. These findings provide reference values for the modification of alpha-amylases.

Which one is important to achieve maximum degree of hydrolysis, starch pre-treatment or activated α-amylase: kinetics and mathematical modeling for liquefaction

Summary: In order to investigate the effectiveness of starch pretreatment and enzyme modification in achieving maximum hydrolysis, corn starch and potato starch were pre-treated using freezing-thawing, ultrasonication, and high-pressure methods. The results showed that pre-treated starches were more susceptible to amylolysis due to the disruption of granule structure and the formation of defects on the surface. The kinetics of starch hydrolysis were evaluated using a first-order equation, and it was found that pressurized potato starch and enzyme activation had the greatest influence on the rate of hydrolysis.

Ultrasound-assisted activation amylase in the presence of calcium ion and effect on liquefaction process of dual frequency ultrasonicated potato starch

Summary: This study aimed to compare the enzymatic digestibility of non-activated and activated amylases with the co-operation of Ca2+ and ultrasonication on potato starch. Ultrasonication caused holes and cracks on the surface of starch, making it more susceptible to amylolysis. Alpha-amylase activated with both sonication and Ca2+ showed higher liquefaction process and could reduce enzyme consumption in various industries.

Ultrasound-Assisted Detoxification of Ochratoxin A: Comparative Study of Cell Wall Structure, Hydrophobicity, and Toxin Binding Capacity of Single and Co-culture Lactic Acid Bacteria

Summary: This study aimed to evaluate the binding capacity and stability of ochratoxin A (OTA) by single and co-culture Lactobacillus acidophilus + Lactobacillus rhamnosus in various modes. It was found that ultrasound-treated L. acidophilus + L. rhamnosus exhibited the highest OTA adsorption capacity and stability. Additionally, the reduction of OTA significantly decreased its toxic performances on cells.
