Iron metabolism and ferroptosis in type 2 diabetes mellitus and complications: mechanisms and therapeutic opportunities

Runyu Miao et al.

Summary: The balance of iron homeostasis is crucial for proper endocrine function, and an imbalance in iron levels has been linked to the development of various endocrine diseases. Ferroptosis, an iron-dependent form of cell death, has emerged as an important process in the pathogenesis and progression of type 2 diabetes mellitus (T2DM). Understanding the mechanisms of iron metabolism and ferroptosis in T2DM may lead to improved disease management. This review discusses the connections between metabolic pathways, molecular mechanisms, and potential treatment targets related to iron metabolism and ferroptosis in T2DM.

Targeting ferroptosis with miR-144-3p to attenuate pancreatic β cells dysfunction via regulating USP22/SIRT1 in type 2 diabetes

Shanshan Zhang et al.

Summary: This study found that Ferroptosis occurred in T2DM and investigated its mechanisms. The up-regulation of miR-144-3p suppressed USP22/SIRT1, leading to Ferroptosis and subsequent dysfunction of pancreatic beta cells, thereby promoting the development of T2DM.

SLC40A1 Mediates Ferroptosis and Cognitive Dysfunction in Type 1 Diabetes

Lijun Hao et al.

Summary: Cognitive dysfunction often accompanies diabetes, and recent evidence suggests that ferroptosis is a key pathogenic pathway involved in this dysfunction, affecting nerve cell death as well as diseases like Alzheimer's, Huntington's, and Parkinson's.

PARP inhibition promotes ferroptosis via repressing SLC7A11 and synergizes with ferroptosis inducers in BRCA-proficient ovarian cancer

Ting Hong et al.

Summary: The study reveals that ferroptosis contributes to the efficacy of PARP inhibitor olaparib by downregulating SLC7A11 expression. Interference with ferroptosis leads to resistance to olaparib, while boosting ferroptosis enhances sensitivity to PARP inhibitors in BRCA-proficient ovarian cancer cells.
