Topical Losartan: Practical Guidance for Clinical Trials in the Prevention and Treatment of Corneal Scarring Fibrosis and Other Eye Diseases and Disorders

Summary: Losartan, an angiotensin II receptor blocker, inhibits TGF beta signaling by blocking ERK activation. Studies have shown its efficacy in reducing scarring fibrosis in various eye injuries and complications. Clinical studies are needed to explore its effectiveness and safety in treating corneal scarring fibrosis and other eye diseases involving TGF beta. It has the potential to enhance pharmacological therapeutics for ocular diseases where TGF beta plays a central role.

Corneal fibroblast collagen type IV negative feedback modulation of TGF beta: A fibrosis modulating system likely active in other organs

Summary: Collagen type IV plays a key role in controlling the pro-fibrotic effects of transforming growth factor beta. It binds TGF beta and regulates its function in various organs. Losartan, a known TGF beta signaling inhibitor, can alter the production of collagen type IV and potentially mitigate the effects of TGF beta. Similar pathways may exist in other organs as well.

Topical Losartan and Corticosteroid Additively Inhibit Corneal Stromal Myofibroblast Generation and Scarring Fibrosis After Alkali Burn Injury

Summary: This study evaluated the efficacy of losartan and prednisolone acetate in inhibiting corneal scarring fibrosis after alkali burn injury in rabbits. The results showed that the combined use of these two drugs significantly reduced opacity area and intensity, decreased myofibroblast area and staining intensity, and confined myofibroblasts to the posterior stroma while promoting repopulation of the anterior and mid-stroma with keratocan-positive keratocytes. Furthermore, topical losartan reduced the production of collagen type IV. These findings suggest that topical losartan and prednisolone acetate can effectively decrease myofibroblast-mediated scarring fibrosis after corneal injury.

TGFβ1 and TGFβ2 proteins in corneas with and without stromal fibrosis: Delayed regeneration of apical epithelial growth factor barrier and the epithelial basement membrane in corneas with stromal fibrosis

Summary: The study aimed to investigate the expression and localization of TGF beta 1 and TGF beta 2 in rabbit corneas with and without stromal fibrosis, as well as defective perlecan incorporation in the epithelial basement membrane. The findings revealed delayed regeneration of epithelial and EBM barriers in high injury corneas, leading to increased myofibroblast development and stromal fibrosis. Corneas with more severe injuries showed prolonged myofibroblast viability and triggered stromal scarring fibrosis due to delayed regeneration of epithelial and EBM barriers.

Descemet's membrane injury and regeneration, and posterior corneal fibrosis, in rabbits

Summary: This study investigated Descemet's membrane and corneal endothelial regeneration, myofibroblast generation and disappearance, and TGF beta-1 localization after Descemetorhexis in rabbits. The results showed that stromal transparency increased over time, myofibroblast zone thickness peaked at one month post-injury and then decreased, and TGF beta-1 deposition peaked at one day post-injury and decreased before regeneration of the endothelium and Descemet's membrane.

Automated Western immunoblotting detection of anti-SARS-CoV-2 serum antibodies

Summary: ELISA and chemiluminescence serological assays for COVID-19 currently focus on one or two SARS-CoV-2 antigens, while an automated Western immunoblotting was developed as a complementary serologic assay, using an inactivated SARS-CoV-2 lineage 20a strain as the antigen source. The automated Western immunoblotting showed substantial agreement (90%) with the compared ELISA, with a sensitivity of 94% and a specificity of 93%, particularly in samples collected more than 2 weeks after symptom onset.

TGF beta-1,-2 and-3 in the modulation of fibrosis in the cornea and other organs

Summary: The TGF beta-1, -2 and -3 isoforms, transcribed from different genes, share the same regulatory mechanisms and signal transduction pathways. Despite the traditional belief that TGF beta-1 and TGF beta-2 are pro-fibrotic while TGF beta-3 is anti-fibrotic, this review suggests that TGF beta-3 actually shares more similarities in regulating fibrotic gene expression with TGF beta-1 and TGF beta-2. The complex interplay between these three isoforms and various proteins controls the healing process in response to injury and the resolution of fibrosis in different organs.

Interleukin-1 and Transforming Growth Factor Beta: Commonly Opposing, but Sometimes Supporting, Master Regulators of the Corneal Wound Healing Response to Injury

Summary: This review highlights the interactions and regulatory roles of interleukin (IL)-1 and transforming growth factor (TGF) beta in the corneal wound healing process. Both systems play crucial roles in regulating cellular functions and gene expressions during the healing response to injuries or infections, and their coordinated downregulation is necessary to terminate the response and prevent fibrosis in the cornea. Understanding the IL-1 and TGF beta systems better can lead to improved approaches for controlling excessive healing responses and scarring corneal fibrosis caused by infections and injuries.
