Lipid-polymer hybrid nanoparticle with cell-distinct drug release for treatment of stemness-derived resistant tumor

Summary: Drug resistance is a major reason for the failure of cancer chemotherapy. Cancer stem-like cells (CSCs), which have high tumorigenicity and innate chemoresistance, can survive traditional chemotherapy and develop increased resistance. This study developed a lipid-polymer hybrid nanoparticle to overcome CSC-associated chemoresistance by co-delivering the differentiation-inducing agent all-trans retinoic acid (ATRA) and the chemotherapeutic drug doxorubicin (DOX). The nanoparticles achieve differential release of the drugs in CSCs and bulk tumor cells, enhancing therapeutic efficacy.

Summary: Although radiotherapy has become an indispensable method for tumor treatment, the side effects and lack of appropriate radiosensitizers limit its further clinical applications. In this study, we synthesized BiOI nanodots coated with M1 macrophage membrane, which enhanced radiosensitivity and could be excreted quickly. The M1 macrophage membrane acted as a carrier with long-circulating and tumor-targeting capabilities. This tumor targeting and renal clearable radiosensitizer improved the therapeutic outcome of radiotherapy and reduced potential toxicity.

Enzyme-triggered deep tumor penetration of a dual-drug nanomedicine enables an enhanced cancer combination therapy

Summary: Emerging nanomedicines can eliminate cancer cells by promoting excessive intracellular reactive oxygen species (ROS) generation. However, tumor heterogeneity and poor penetration of nanomedicines often result in varying levels of ROS production, where low levels of ROS can actually promote tumor cell growth, reducing the effectiveness of these nanomedicines. In this study, a novel amphiphilic and block polymer-dendron conjugate-derived nanomedicine (GFLG-DP/Lap NPs) was developed, incorporating a photosensitizer (Ppa) for ROS therapy and Lapatinib (Lap) for molecular targeted therapy. By combining ROS therapy with Lap, the synergistic effect effectively kills cancer cells, even in deep tumor regions.

Microbiotic nanomedicine for tumor-specific chemotherapy-synergized innate/adaptive antitumor immunity

Summary: This study reports a novel microbiotic nanomedicine LOD/TPZ@Lips-LA, which combines lactate oxidase (LOD) and prodrug tirapazamine (TPZ) loaded liposomes anchored on lactobacillus (LA) surface for chemotherapy-synergized antitumor immunity. The nanomedicine induces tumor cell apoptosis through in situ ROS generation and TPZ activation based on LA metabolism, resulting in strong immunogenic cell death (ICD). By regulating biomarker expression, it promotes anti-tumorigenic M1 polarization of innate tumor-associated macrophages and infiltration of adaptive cytotoxic CD8+ T cells, synergistically enhancing the tumor inhibition effects.
