Prevalence, Risk, and Challenges of Extemporaneous Preparation for Pediatric Patients in Developing Nations: A Review

Sri Hartati Yuliani et al.

Summary: Extemporaneous preparations are widely used for pediatric patients with special treatments, but they come with several problems that can lead to adverse events or a lack of therapeutic effectiveness. Developing nations are facing challenges in compounding practices, and the prevalence of compounded medication in these nations needs to be explored to determine the urgency of addressing these challenges. Through investigation and collection of scientific articles from reputable databases, the risks and challenges associated with extemporaneous preparations are described and explained.

KRAS inhibition in metastatic colorectal cancer: An update

Maliha Nusrat et al.

Summary: Approximately 50% of colorectal cancers have mutations in the KRAS gene, which are associated with worse prognosis and limited targeted therapy options. This review discusses clinical approaches to target KRAS in colorectal cancer, focusing on downstream inhibitors and recent direct inhibitors for KRASG12C and other mutants. Early clinical trial data indicates limited activity of these novel inhibitors compared to other cancer types, and the role of receptor tyrosine kinase signaling and parallel signaling pathways in modulating response to these inhibitors is explored. Additionally, the review examines the impact of KRAS mutations on the tumor-immune microenvironment and efforts to induce an immune response against these tumors.

Associating chitosan and microemulsion as a topical vehicle for the administration of herbal medicines

Denise A. J. Oliveira et al.

Summary: By associating low molecular weight chitosan with a microemulsion, a more viscous liquid crystal topical vehicle was formed. Experimental results with Brazilian Stryphnodendron adstringens shell extract and Melaleuca alternifolia essential oil showed activity against Candida albicans. Incorporating chitosan in the microemulsion improved the mechanical properties of the topical formulation, providing advantages for the topical treatment of vulvovaginal candidiasis.

The loss of SHMT2 mediates 5-fluorouracil chemoresistance in colorectal cancer by upregulating autophagy

Jian Chen et al.

Summary: This study identified SHMT2 as a critical regulator of 5-FU chemoresistance in colorectal cancer (CRC), with SHMT2 inhibiting autophagy and promoting apoptosis under 5-FU treatment. Autophagy inhibitors could reduce 5-FU resistance induced by low SHMT2 levels. Targeting the SHMT2-p53 interaction may present a novel therapeutic strategy to overcome chemoresistance in CRC.

Microemulsion for Prolonged Release of Fenretinide in the Mammary Tissue and Prevention of Breast Cancer Development

Giovanna Cassone Salata et al.

Summary: The study developed a nonaqueous microemulsion capable of forming a depot in mammary tissue for prolonged release of fenretinide to prevent breast cancer development. The microemulsion released 30% of fenretinide in vitro and reduced cell migration and spheroid growth in cell cultures. In a preclinical model, microemulsion administration significantly reduced breast tumor incidence and increased type II collagen expression, showing potential as a preventive therapy for breast cancer.

Influence of Blending of Nonionic Emulsifiers Having Various Hydrophilic Head Sizes on Lipid Oxidation: Investigation of Antioxidant Polarity-Interfacial Characteristics Relationship

Jiyun Lee et al.

Summary: This study aimed to investigate the effects of interfacial composition and antioxidant polarity on lipid oxidation of emulsions. Results showed that the hydrophilic head size and concentration of surfactants can affect the rate of lipid oxidation, but not the antioxidant ability. Mixing surfactants with different hydrophilic head sizes and decreasing surfactant concentration enhanced the ability of antioxidants to prevent lipid peroxide generation. Additionally, lauryl gallate exhibited a more effective antioxidant ability than TBHQ in most emulsions.

Effect of Bile Salts on the Interfacial Dilational Rheology of Lecithin in the Lipid Digestion Process

Aicha Mekkaoui et al.

Summary: The effects of bile salts on the emulsifier adsorption layer are crucial in lipid digestion. The rapid diffusion of NaCh molecules can break the emulsifier interface layer, enhancing the viscoelasticity of the interface film and accelerating material exchange between the bulk and interface, facilitating lipase adsorption and promoting digestion. Understanding the interfacial behavior of bile salts with emulsifiers provides a basis for modulating lipid digestion through rational design of the interfacial layer.
